Tumor necrosis factor alpha, also called cachectin, is the best-know member of the TNF-family, which can cause cell death. This protein is produced by neutrophils, activated lymphocytes, macrophages, NK cells, LAK cells, astrocytes endothelial cells, smooth muscle cells and some transformed cells. TNF-α occurs as a secreted, soluble form and as a membrane-anchored form, both of which are biologically active. The naturally-occurring form of TNF-α is glycosylated, but non-glycosylated recombinant TNF-α has comparable biological activity. The biologically active native form of TNF-α is reportedly a trimer. Human and mouse TNF-α show approximately 79% homology at the amino acid level and cross-reactivity between the two species.

  • We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute in sterile ultrapure water or aqueous buffer containing 0.1% BSA to a concentration of 0.1-1.0mg/ml. Stock solutions should be apportioned into working aliquots and stored at ≤-20ºC. Further dilutions should be made in appropriate buffered solutions. If necessary, perform 0.2μm filtration.

Storage Conditions

Store at −20°C or below. The product is stable for at least one year under the recommended storage conditions.

 

Precautions

  • Each freeze–thaw cycle may result in partial loss of protein activity. After initially reconstituting the product to the appropriate concentration (please refer to the Reconstitution section of the product information for the recommended formulation), aliquot the solution and store at −20°C or below to minimize repeated freeze–thaw cycles.
  • Certain proteins may exhibit strong adsorption to the surfaces of some plastic tubes, resulting in protein loss and potentially irreversible binding to the tube wall. Carrier proteins, such as 0.1% BSA, can block protein-binding sites on plastic surfaces and reduce nonspecific adsorption. Therefore, it is strongly recommended to prepare the stock solution according to the instructions provided in the Reconstitution section of the product information. Failure to do so may result in product loss due to adsorption to the tube wall.
  • Although the solution has been sterilized prior to lyophilization, it is recommended to filter-sterilize the reconstituted solution through a 0.2 μm membrane filter before use in cell culture or other downstream applications. Maintain aseptic technique throughout handling and use.
  • This product is intended for professional scientific research use only. It is not intended for clinical diagnosis or treatment, and must not be used in food or pharmaceutical products. Do not store in ordinary residential premises.
