MCA-EIDLMVLDK-Dnp is a fluorogenic substrate for β-secretase 1 (β-site amyloid cleavage enzyme 1, BACE1). Its mechanism is based on fluorescence resonance energy transfer (FRET). After cleavage, this substrate releases the fluorescent group MCA (7-methoxycoumarin-4-yl)acetyl, with a maximum excitation wavelength of 320 nm and maximum emission wavelength of 405 nm. By measuring fluorescence, BACE1 enzyme activity can be detected. The cleavage rate of this substrate by BACE1 is approximately 13 times faster than that of the common Swedish variant peptide (EVNLDAEF). Therefore, this substrate is widely used in screening for BACE1 inhibitors or in fluorescent detection of BACE1 activity. MCA: (7-methoxycoumarin-4-yl)acetyl, Dnp: 2,4-dinitrophenyl.
