Description
GenoJump™ pG-Tn5 Transposase is an engineered fusion protein derived from E. coli, consisting of a highly active mutant Tn5 transposase and Protein G. After assembly with appropriate sequencing adapters, pG-Tn5 Transposase becomes the pG-Tn5 Transposome, which can be used for CUT&Tag experiments.
CUT&Tag (Cleavage Under Targets and Tagmentation) is an emerging method for studying protein-DNA interactions. It offers high specificity, low background noise, high sensitivity, good reproducibility, and low cost, making it a suitable replacement for the traditional ChIP-Seq (Chromatin Immunoprecipitation-sequencing) method. The core of CUT&Tag technology lies in the ability of Protein A or Protein G on the pA/G-Tn5 Transposome to specifically bind to antibodies targeting the protein of interest. This allows the Tn5 transposome to form a complex with the target protein via Protein A/G and the antibody. The addition of Mg2+ activates the enzyme activity of the Tn5 transposome, which cleaves the DNA bound to the target protein, achieving targeted DNA fragmentation. Simultaneously, the sequencing adapters carried by the transposome are attached to the ends of the DNA fragments, enabling tagging. Subsequent PCR reactions fill in the gaps and amplify the DNA fragments using dual-indexed PCR primers, preparing a DNA library suitable for next-generation sequencing (NGS) platforms.
GenoJump™ pG-Tn5 Transposase is a recombinant protein expressed and purified using our ExtremeProtein™ technology platform. Compared to wild-type Tn5 transposase, it offers at least 1000 times higher insertion efficiency, along with better insertion randomness, high stability, and easy sequencing of insertion sites.
Enzyme Storage Solution
50mM HEPES (pH7.2), 100mM NaCl, 0.1mM EDTA, 1mM DTT, 0.1% Triton X-100, 50% (v/v) Glycerol.
Precautions
- This product contains 50% glycerol and will not freeze when stored at -20°C. Avoid storage at -80°C, as it will freeze under those conditions. Repeated freeze-thaw cycles may reduce the enzyme activity of this product.
- The product is relatively viscous, so be careful to ensure accurate sampling. After adding the sample, mix thoroughly by pipetting up and down to avoid the formation of bubbles.
- This product is intended for scientific research by professionals only. It is not to be used for clinical diagnostics or treatments, food, or pharmaceuticals, and should not be stored in ordinary residences.
- For your safety and health, please wear a lab coat and disposable gloves when handling this product.
Storage
The minimum shelf life is 1 year at -20°C.
