Description
Albumin/IgG Depletion Kit, also known as the Albumin/IgG Removal Kit, is an efficient and convenient reagent kit designed for the rapid removal of albumin and Immunoglobulin G (IgG) from serum, plasma, or other samples using a mixture of specific Cibacron Blue dye-agarose gel and Protein A/G agarose gel. Samples depleted of albumin and IgG are commonly used in the preparation of protein samples for techniques such as Western blotting, enzyme-linked immunosorbent assay (ELISA), immunoassays, 2D electrophoresis, or mass spectrometry research.
The kit exhibits some effectiveness in removing human IgA, IgD, IgE, and IgM, but the removal efficiency for mouse and rat IgM is minimal. This characteristic is related to the specificity of the binding of different species' immunoglobulins to Protein A/G.
Proteins in blood include albumin, globulin, and fibrinogen, with serum being the plasma after the removal of fibrinogen. Albumin, also known as clear protein, has a molecular weight of approximately 66 kDa and plays various roles in the body, including maintaining osmotic pressure, providing nutrition, detoxification, and transportation. Serum and plasma are crucial samples for studying proteinomics in humans and other animals. However, these samples are complex and often contain a large amount of albumin (constituting 50%-70% of total serum/plasma proteins) and immunoglobulins (10%-20% of total serum/plasma proteins), with Immunoglobulin G (IgG) being the major form, constituting approximately 75% of the total immunoglobulin content in serum. IgG is primarily involved in recognizing and clearing pathogens and toxic substances, contributing to functions such as antiviral, antibacterial, and immune regulation. High levels of albumin and immunoglobulins can interfere with research by masking low-abundance target proteins, leading to a significantly low detection rate of low-abundance proteins, affecting various analyses, including proteomic studies. Therefore, the removal of albumin and immunoglobulins and the enrichment of target proteins are essential steps in the preparation of blood protein samples.
Cibacron Blue dye, also known as Cibacron Blue 3G-A, or Reactive Blue, is an anionic anthraquinone dye with a spatial structure similar to NAD. It exhibits an affinity adsorption effect with various kinases, dehydrogenases, albumin, DNA polymerase, etc. The covalent binding of Cibacron Blue dye to agarose resin forms a bioaffinity chromatography medium called Blue Gel Agarose or Blue Gel, which is an ideal choice for the removal of albumin and enzyme purification. Recombinantly modified Protein A and Protein G can specifically bind to the Fc end of most mammalian IgGs, with molecular weights of 14 kDa and 22 kDa, respectively. Each Protein A molecule and Protein G molecule can bind to 2 and 3 IgG molecules, respectively. Recombinantly modified Protein A and Protein G, covalently bound to agarose gel, can be used for antibody purification or immunoprecipitation and are also suitable for the removal of immunoglobulins, especially IgG.
The kit is based on Binding Spin Columns filled with a mixture of Blue Gel and Protein A/G agarose gels, allowing the binding of albumin and IgG from the sample.

- This kit offers a high binding capacity. The gel mixture used in this kit is stored in a special protective solution (without glycerol) and is packed into the Binding Spin Column. In comparison to similar kits, the Albumin/IgG Binding Spin Column provided by this kit has a high binding capacity for both albumin and IgG, capable of binding more than 3mg of albumin and more than 0.6mg of IgG per column.
- The operation of this kit is simple, convenient, and rapid. The entire experiment can be completed within 30 minutes through straightforward pipetting and centrifugation steps, enabling the efficient removal of albumin from the sample.
- This kit has a broad application range. It can be used for the removal of albumin and IgG from serum, plasma, antibodies, and other samples, including species such as humans, cows, mice, goats, rabbits, pigs, etc.
