Description
Albumin Depletion Kit for Serum or Plasma is an efficient, convenient, and rapid reagent kit designed for the rapid removal of albumin from serum, plasma, or other samples using specific Cibacron Blue dye-agarose resin. Samples depleted of albumin are commonly used in protein sample preparation for techniques such as Western blotting, enzyme-linked immunosorbent assay (ELISA), immunoassays, 2D electrophoresis, or mass spectrometry research.
Proteins in blood include albumin, globulin, and fibrinogen, among others, with serum being the plasma after the removal of fibrinogen. Albumin, also known as clear protein, has a molecular weight of approximately 66 kDa and plays roles in maintaining osmotic pressure, providing nutrition, detoxification, and transportation. Serum and plasma are crucial samples for studying proteinomics in humans and other animals. However, these samples are complex, often containing a large amount of albumin, constituting 50%-70% of total serum/plasma proteins. High levels of albumin can be an interference factor in research, masking low-abundance target proteins and leading to a significantly low detection rate of low-abundance proteins, thereby affecting various analyses, including proteomic studies. Therefore, removing albumin and enriching target proteins are essential steps in the preparation of blood protein samples.
Cibacron Blue dye, also known as Cibacron Blue 3G-A, or Reactive Blue, is an anionic anthraquinone dye with a spatial structure similar to NAD. It exhibits an affinity adsorption effect with various kinases, dehydrogenases, albumin, DNA polymerase, etc. This affinity is attributed to the structural similarity between the dye and the natural ligands of proteins with cofactor-binding domains. The covalent binding of Cibacron Blue dye to agarose resin forms a bioaffinity chromatography medium called Blue Gel Agarose or Blue Gel, which is an ideal choice for the removal of albumin and enzyme purification. It is widely used in binding and separating albumin and protein purification in blood samples, as well as exosome purification.
The kit is based on Binding Spin Columns filled with Blue Gel Agarose, allowing the binding of albumin from the sample. Removal of albumin from the sample is achieved through centrifugation.
