Description
The DNA mismatch repair (MMR) system primarily consists of three core proteins: MutS, MutL, and MutH. MutS recognizes and binds to mismatched bases or unpaired DNA sites, followed by the recruitment of MutL to form a stable MutS-MutL-DNA complex. This complex further activates the endonuclease activity of MutH, which cooperates with helicases and single-strand DNA-binding proteins to remove erroneous mismatch sequences.
Aae MutS is a thermostable mismatch repair protein derived from the thermophilic bacterium Aquifex aeolicus. It exhibits DNA-dependent ATPase activity within its optimal reaction temperature range of 65–75 °C. During DNA replication, Aae MutS specifically recognizes mismatched base pairs and loop structures formed by insertions. By tightly binding these erroneous structures, Aae MutS effectively blocks DNA polymerase extension and suppresses error-prone amplification.
Aae MutS binds mismatched DNA with 10–20-fold higher affinity than correctly paired DNA, showing the strongest binding preference for G/T mismatches. This unique property enables broad applications in ARMS-PCR, microarray-based genotyping, and gene synthesis. It efficiently eliminates mismatched sequences, reduces mutation rates, and improves the fidelity of synthetic gene products.
Our recombinant AaeMutS protein is heterologously expressed in E. coli and purified via affinity chromatography. It exhibits excellent thermostability with a half-life longer than 40 minutes, fully compatible with high-temperature PCR workflows.
