5’-Phosphate Dependent Exonuclease (XRN-1)

XRN-1, also known as 5’-Phosphate Dependent Exonuclease, is a highly active 5’-3’ exoribonuclease expressed and purified from Escherichia coli. It requires the substrate RNA to have a 5’ monophosphate and catalyzes the degradation of 5’ monophosphate RNA (p-RNA) into NMP in the 5’→3’ direction. XRN-1 can also digest single-stranded DNA (ssDNA) with a 5’ monophosphate, though the cleavage efficiency is lower. XRN-1 cannot digest RNA with 5’ triphosphate (5’-ppp), 5’ diphosphate (5’-pp), 5’ cap, or 5’ hydroxyl (5’-OH), as well as double-stranded DNA (dsDNA) or ssDNA with 5’ hydroxyl, triphosphate, or diphosphate. XRN-1 also cannot digest 5’ phosphate-ended recessed double-stranded RNA, DNA-RNA hybrids, or RNA in secondary structures.

Description

XRN-1, also known as 5’-Phosphate Dependent Exonuclease, is a highly active 5’-3’ exoribonuclease expressed and purified from Escherichia coli. p-RNA → NMP

The enzymatic properties of XRN-1 make it useful for selectively digesting ribosomal RNA (rRNA) to isolate and enrich messenger RNA (mRNA) from total RNA without relying on oligo-dT magnetic beads or agarose gels, facilitating the construction of mRNA sequencing libraries. XRN-1 can also be used for 5’ end RNA analysis.

Expressed in Escherichia coli, with the gene encoding XRN-1 from yeast.

Approximately 175 kDa.

 

Activity Definition

One unit digests 1 µg of rRNA substrate into acid-soluble nucleotides in 60 minutes at 30ºC under standard assay conditions.

 

Purity

Free from RNAse and DNA endonucleases and exonucleases other than its own activity.

  • During use, it is recommended to keep the product in an ice box or on an ice bath. After use, it should be immediately stored at -20°C.
