Description

2×Taq Plus PCR Mix (With Dye) is a pre-mixed PCR solution formulated with Taq DNA Polymerase. It includes reaction buffer, dNTPs, and other components, making it convenient and quick to use while reducing contamination during PCR procedures. Simply take an appropriate amount of 2×Taq Plus PCR Mix (With Dye), add the template and primers, and supplement with ddH2O to achieve a 1× concentration for the PCR reaction.

This mix can amplify DNA fragments up to 5 kb in length, offering excellent amplification specificity and template compatibility. The PCR products have an A overhang at the 3’ end, making them suitable for direct T/A cloning after purification.

The PCR mix contains two dyes, allowing direct loading of PCR products onto gels without the need for additional loading buffer. During electrophoresis, two indicator bands will appear. These dyes do not affect PCR amplification efficiency, but for experiments requiring optical analysis of PCR products (e.g., absorbance, fluorescence), it is recommended to purify the PCR products before analysis or use the dye-free 2×Taq Plus PCR Master Mix.

Quality Control

Nuclease Activity Test
25 μl of 2×Taq Plus PCR Mix (With Dye) is mixed with 200 ng of supercoiled plasmid DNA to form a 50 μl reaction system. After incubation at 37°C for 4 hours, agarose gel electrophoresis is used to detect that less than 10% of the plasmid DNA has converted to nicked or linear forms.

Non-Specific Nuclease Activity Test
25 μl of 2×Taq Plus PCR Mix (With Dye) is mixed with 15 ng of double-stranded DNA fragments to form a 50 μl reaction system. After incubation at 37°C for 16 hours, agarose gel electrophoresis is used to confirm that the double-stranded DNA substrate remains unchanged.

Storage

The minimum shelf life is 1 year at -20°C.
