Description
β1-4 Galactosidase, also known as β(1→4) galactosidase, E. coli β(1→4) galactosidase, or simply β-galactosidase, β-gal, or lacZ, is a recombinant exoglycosidase derived from the Escherichia coli(E. coli) lacZgene. It catalyzes the hydrolysis of lactose into its constituent monosaccharides.
This enzyme is a homotetramer and requires magnesium (Mg²⁺) and sodium (Na⁺) ions as cofactors for optimal activity. β1-4 Galactosidase specifically hydrolyzes lactose into galactose and glucose. In addition to its hydrolytic function, it exhibits transglycosylation activity, enabling the conversion of lactose into allolactose, which can subsequently be hydrolyzed into galactose and glucose. Notably, β1-4 galactosidase plays a central role in the Lac operon regulatory system in E. coli.
Two commonly used synthetic substrates for β1-4 galactosidase are:
X-Gal (5-Bromo-4-chloro-3-indolyl β-D-galactopyranoside): used for qualitative blue-white screening in molecular biology.
ONPG (o-Nitrophenyl β-D-galactopyranoside): used for quantitative spectrophotometric assays of enzymatic activity.
