
Dual-Luciferase Reporter Gene Detection Service
Description
The Dual-Luciferase Reporter Assay first uses luciferin as a substrate to detect Firefly luciferase, then uses coelenterazine as a substrate to detect Renilla luciferase. When coelenterazine is added, a substance that inhibits the catalytic activity of Firefly luciferase on luciferin is also added, quenching more than 99.9% of the Firefly luciferase signal. This ensures that only the activity of Renilla luciferase is detected in subsequent steps, achieving a dual-luciferase reporter gene detection.
Typically, the transcriptional regulatory elements or 5' promoter region of the gene of interest are cloned upstream of the luciferase, or the 3' UTR region is cloned downstream, constructing a reporter gene plasmid. Cells are then transfected, treated with appropriate agents, lysed, and the luciferase activity is measured. The level of luciferase activity indicates the transcriptional regulatory effects of the treatment on the target gene. Renilla luciferase is often used as an internal control for transfection efficiency to eliminate differences in cell number and transfection efficiency.
