Western Blot Enhancer is a reagent that enhances the specific binding between antigen and antibody during Western blot detection by directly diluting it with primary or secondary antibodies. It can significantly increase the sensitivity of Western blot detection by several-fold to more than ten-fold.
This product is derived from large-scale and fine screening to obtain high-molecular compounds that likely interact specifically with antigen-antibody complexes through adhesion, entanglement, or binding. It also includes complementary blocking reagents and detergents to reduce background, effectively addressing common issues encountered in Western blotting, such as low sensitivity and high background. It greatly improves the signal-to-noise ratio. This product can also be used for enhancing immunofluorescence, immunohistochemistry, dot blot, ELISA, and other immunodetection methods involving antigen-antibody interactions. It can also be attempted for enhancing other protein-protein interaction assays.
Western blot, also known as Western blotting or immunoblotting, often abbreviated as WB, is one of the main methods for protein expression analysis using antibodies. The typical steps involve SDS-PAGE electrophoresis, membrane transfer, blocking, primary antibody incubation, secondary antibody incubation, and detection using labeled secondary antibodies.
During Western blot experiments, common issues encountered include weak signal bands due to low abundance of the target protein or low antibody potency, high background due to poor blocking, and non-specific bands caused by poor antibody specificity. - During the actual use of this product, if the signal of the target band is very strong, the signal of non-specific bands may also be enhanced accordingly. If the non-specific bands are too close to the target protein band and affect the detection results, you can try reducing the protein loading amount, lowering the concentration of primary and secondary antibodies, or shortening the exposure time.
