Description
WELQ Protease exhibits specificity towards the tetrapeptide sequence Trp-Glu-Leu-Gln (W-E-L-Q), selectively cleaving peptide bonds following Gln (Q). It is commonly used for the removal of histidine (His) or other fusion tags from recombinant proteins, with the notable advantage of leaving no additional amino acid residues at the N-terminus of the target protein after cleavage.
The reaction conditions for WELQ Protease are relatively broad. It is a serine protease-like protein derived from Staphylococcus aureus. This protease does not require specific reaction buffers and can recognize the WELQ peptide sequence over a wide temperature range (4-30°C) and pH range (6.5-9.0), facilitating protein or peptide cleavage after the Gln (Q) residue.
WELQ Protease is suitable for on-column cleavage of His-tagged proteins. With its own His-tag, WELQ Protease is particularly suitable for on-column cleavage of proteins tagged with histidine. During the cleavage of His-tagged proteins, the cleaved His tag and WELQ Protease can bind to a His-tag purification resin column, while the target protein remains in the flow-through solution. This enables the elution of the target protein without theoretically containing the His tag and WELQ Protease, greatly facilitating the purification process. If on-column cleavage is not performed, WELQ Protease and the cleaved tag from the target protein can be conveniently removed separately through nickel affinity chromatography and purification columns designed to remove fusion tags.
WELQ Protease has a molecular weight of approximately 22kDa and a purity of ≥95%. It does not contain DNA endonucleases or exonucleases, RNAse, or phosphatases.
The enzymatic activity unit is defined as the amount of enzyme required to cleave ≥99% of 100µg of a control protein in 16 hours at 20°C.
The storage buffer composition for WELQ Protease is as follows: 10mM Na2HPO4, 1.8mM KH2PO4, 140mM NaCl, 2.7mM KCl, 50% (v/v) Glycerol, pH 7.3.
The WELQ Protease cleavage system is compatible with 0.01-1% Triton X-100/Tween-80, 5-50mM DTT, 100mM NaCl, 20-300mM Imidazole, 100mM Tris-HCl (pH 8.0), 100mM Na3PO4 (pH 7.4), and 20mM K3PO4 (pH 7.4).
Precautions
- 2M urea and 0.25M guanidine hydrochloride can partially inhibit the activity of WELQ Protease.
