Description
Tth Nucleoside Diphosphate Kinase is a thermally stable nucleoside diphosphate kinase originating from the thermophilic bacterium Thermus thermophilus. This enzyme catalyzes the transfer of high-energy phosphate groups between adenosine triphosphate (ATP) and nucleoside diphosphates (NDP). Additionally, it possesses NDP kinase activity and protein phosphorylation activity, playing a role in transcription regulation and signal transduction. This enzyme exhibits excellent thermostability and is responsible for synthesizing ribonucleoside triphosphates (rNTPs) other than ATP within cells, converting ribonucleoside diphosphates (rNDPs) into ribonucleoside triphosphates (rNTPs): (d)XDP+(d)YTP→(d)XTP+(d)YDP. The specific activity of this enzyme is 1×105 U/mg, and its packaging concentration is approximately 100 ng/μl. Activity Definition
One unit of activity is defined as the amount of enzyme required to convert 1.0 μmole of UDP to UTP per minute at 65°C, in a reaction buffer system containing 1× Nucleoside diphosphate kinase buffer, 10mM ATP, and 1mM UDP.
Activity Assay Conditions
1× NDPK Buffer: 20 mM HEPES, pH 7.5, 100 mM Potassium glutamate, 10 mM Magnesium acetate, 5mM DTT.
