Description
Tth Endonuclease IV, also known as thermostable Endonuclease IV, is derived from Thermus thermophilus. As a DNA damage repair enzyme, Tth Endonuclease IV participates in the repair of various oxidative DNA lesions.
This enzyme recognizes apurinic/apyrimidinic (AP) sites on double-stranded DNA and cleaves the phosphodiester bond at the 5′ side of AP sites, generating 3′-hydroxyl and 5′-deoxyribose phosphate (5′-dRP) termini. Tth Endonuclease IV also possesses 3′-diesterase activity, which removes 3′-phosphate, 3′-α,β-unsaturated aldehyde (UA), phosphoglycoaldehyde and other 3′ blocking moieties from damaged double-stranded DNA ends. In addition, it exhibits 3′→5′ exonuclease activity.
Tth Endonuclease IV does not require Mg²⁺ or Mn²⁺ for catalytic activity; nevertheless, supplementation with Mg²⁺ or Mn²⁺ can enhance its enzymatic activity.

Unit Definition
One unit is defined as the quantity of enzyme needed to cleave 1 pmol of a 60-mer double-stranded oligonucleotide containing a single AP site in a total reaction volume of 10 μL after incubation at 65 °C for 1 hour.
Purity
Free of all other DNA endonucleases, exonucleases and RNases except Tth Endonuclease IV.
Inactivation & Inhibition
This enzyme cannot be inactivated by heating. Tth Endonuclease IV can be fully inactivated by adding EDTA to a final concentration of 10 mM.
Storage Conditions
Store at -20 °C, stable for at least 2 years.
