Tte UvrD Helicase

Tte UvrD Helicase is derived from the thermostable bacterium Thermoanaerobacter tengcongensis uvrD gene. The protein has the ability to unwind 3 '- 5' DNA under thermostable conditions. In the isothermal amplification at high temperature [thermophilic Helicase Dependent Amplification (tHDA)], this enzyme can assist the polymerase in strand unwinding, thus completing the isothermal amplification. The enzyme is resistant to 65°C and can be used for isothermal amplification independent of the single-strand binding protein.

Description

Tte UvrD Helicase is derived from the thermostable bacterium Thermoanaerobacter tengcongensis uvrD gene. The protein has the ability to unwind 3 '- 5' DNA under thermostable conditions. In the isothermal amplification at high temperature [thermophilic Helicase Dependent Amplification (tHDA)], this enzyme can assist the polymerase in strand unwinding, thus completing the isothermal amplification. The enzyme is resistant to 65°C and can be used for isothermal amplification independent of the single-strand binding protein.

 

Components

2 μg

Tte UvrD helicase (20 μg/ml):    100 μl

50 μg
Tte UvrD helicase (1 mg/ml):    50 μl

 

Applications

  • DNA unwinding in isothermal amplification.

 

Storage

The minimum shelf life is 3 years at -20°C.

 

Note

  • 3mM ATP and 2mM Mg2+should be added to the enzyme reaction solution.

  • It is reported in the literature that adding Tte UvrD Helicase 4ng/μl to the tHDA reaction system can realize tHDA amplification, which does not depend on Tte MutL single-strand binding protein.
