Description
Tte UvrD helicase, derived from thermophilic bacteria, is capable of unwinding DNA under heat-stable conditions. Our tests have confirmed that this helicase can efficiently unwind 5'-overhang, 3'-overhang, and blunt-ended double-stranded DNA. The unwinding activity of this enzyme depends on auxiliary factors ATP or dATP. The Tte UvrD helicase remains active within a temperature range of 45–65°C, with enzyme inactivation occurring at temperatures exceeding 65°C. With its exceptional unwinding activity, this enzyme holds potential for applications such as NanoPore sequencing, chip hybridization, or tHDA amplification, although these functionalities have not been tested by us.

Storage
The minimum shelf life is 3 years at -20°C.
Notes
- The reaction mixture for this enzyme requires the addition of 3mM ATP or dATP and >2mM Mg²⁺.
- The enzyme's functionality testing 1x Buffer composition is as follows: 20mM Tris-HCl (pH 7.6), 50mM KCl, 10mM MgCl₂, and 3mM ATP. Due to variations in applications, this product does not include the reaction buffer.
- In a 20μl reaction system, combine 2–5 pmol dsDNA with 10–20 ng of this product. The recommended reaction temperature is 60°C for 15–30 minutes. However, adjustments to experimental parameters may be required based on substrate length or type.
