Description
Total Glutathione Assay Kit is a simple and convenient assay kit used to detect the total glutathione (GSSG+GSH) levels.
Glutathione is a small peptide composed of three amino acid residues. Its full name is glutamyl-cysteinyl-glycine, and it is commonly referred to as glutathione. Due to the thiol group (SH) on the cysteine residue, glutathione is often abbreviated as G-SH or GSH. Glutathione exists in two forms: reduced glutathione (GSH) and oxidized glutathione disulfide (GSSG). GSSG is formed from two molecules of GSH through the oxidation of the thiol group and is often abbreviated as G-S-S-G or GSSG. GSH is the primary source of thiol groups in most viable cells and plays a crucial role in maintaining the appropriate redox state of proteins. It is also a key antioxidant in animal cells. Typically, 90-95% of total glutathione is in the reduced form (GSH).
Total Glutathione Assay Kit utilizes glutathione reductase to reduce oxidized glutathione (GSSG) to reduced glutathione (GSH). GSH then reacts with the colorimetric substrate DTNB to produce a yellow-colored TNB and GSSG. By appropriately setting up the reaction system and combining the two reactions, the total glutathione (GSSG+GSH) becomes the rate-limiting factor for color development, and the amount of total glutathione determines the formation of yellow TNB. Therefore, the total glutathione level can be calculated by measuring the absorbance at 412 nm (A412).
This assay kit can be used to detect the total glutathione content in animal tissues, plasma, red blood cells, and cultured cells or other appropriate samples.
The assay kit provides a protein removal reagent (reagent S) to more accurately measure the total glutathione content in samples containing proteins.
The detection limit of this assay kit is 1 μM. One kit allows for 100 tests.
Additional Notes
- During the detection process of this assay kit, oxidation-reduction reactions are involved, and all oxidizing agents or reducing agents can interfere with the measurement. Specifically, reagents containing thiols, such as DTT and mercaptoethanol, can severely interfere with the assay. Please try to avoid using them.
- It is crucial to strictly control the temperature and reaction time during the assay. Otherwise, a standard curve needs to be prepared for each test.
- Reagents like NADPH are not very stable, so it is important to follow the instructions carefully to prevent inactivation.
- DMSO may solidify at lower temperatures, such as 4°C or during ice baths. It can be gently warmed in a water bath at 20-25°C until completely melted before use.
- This product is intended for scientific research purposes by professionals only and is not to be used for clinical diagnosis or treatment. It is not intended for use in food or drugs and should not be stored in a regular residential setting.

Storage
Store at -20°C. Shelf life is one year. Once reduced glutathione is prepared as a solution, it should be appropriately aliquoted and stored at -20°C for a minimum of 3 months. After dissolving DTNB in DMSO, it should be appropriately aliquoted and stored at -20°C for a minimum of 3 months. Once reagent S for protein removal is prepared as a solution, it can be stored at 4°C. After dissolving NADPH, it should be appropriately aliquoted and stored at -70°C.
