Tli (exo-) DNA Polymerase, also known as Vent (exo-) DNA polymerase, is derived from Thermococcus litoralis. It is genetically engineered to remove the proofreading activity of 3´ - 5´ exonuclease, and its fidelity is reduced, about 2 times higher than Taq DNA polymerase. After incubation at 95°C for 1 hour, the enzyme still has more than 90% polymerase activity. Tli (exo-) DNA Polymerase and Vent (exo-) DNA Polymerase have the same properties and use methods. This polymerase is purified from recombinant E.Coli strain, which carries D141A/E143A mutation.

One unit is the amount of enzyme required to catalyze the incorporation of 10 nmol dNTP into acid-insoluble substances within 30 minutes at 74°C.
