Description
Thermostable RecJ Nuclease (3' to 5’) is derived from thermophilic archaea, differing from bacterial RecJ nucleases. This enzyme acts on single-stranded DNA (ssDNA) and single-stranded RNA (ssRNA), hydrolyzing them in the 3´→5´ direction to produce 3’-OH ssDNA, ssRNA, and mononucleotides. It exhibits higher hydrolytic activity on ssRNA compared to ssDNA substrates. The enzyme is highly thermostable, with a half-life of 15 minutes at 95°C, and maintains high activity between 60-85°C. It has low tolerance to NaCl, with concentrations above 50 mM severely inhibiting its activity.
Unit Definition
One unit is defined as the amount of enzyme required to hydrolyze ssDNA to produce 50 pmol of acid-soluble deoxyribonucleotides within 30 minutes at 50°C under 1× RecJ Nuclease reaction buffer conditions.
