Description
Thermostable Flap Endonuclease 1 (FEN1) catalyzes the excision of DNA and RNA flap structures at the junction of double-stranded nucleic acids, generating a 5´-phosphate end. In vivo, FEN1 is an essential component of Okazaki fragment processing and participates in base excision repair processes. Our Thermostable FEN1 is recombinantly expressed and purified through multiple steps.
Activity Definition
1 unit is defined as the amount of enzyme required to cleave 10 pmol of oligonucleotide substrate containing a 5´ flap structure (20 nt flap) at 65°C for 10 minutes.
Activity Assay Conditions
1× AM Buffer E: 20 mM Tris-HCl, 10 mM (NH4)2SO4, 10 mM KCl, 10 mM MgSO4, 0.1% Triton X-100 (pH 8.8 @ 25°C), incubation at 65°C.
