Description
Thermostable Endonuclease Q, also referred to as EndoQ or Endo Q, is a DNA endonuclease derived from archaea. This enzyme recognizes and cleaves the phosphodiester bond immediately upstream of deoxyxanthosine (dX), deoxyinosine (dI), deoxyuridine (dU) and abasic (AP) sites, generating a nick with a 3'-hydroxyl terminus and a 5'-phosphate terminus after cleavage. Thermostable Endonuclease Q exhibits higher cleavage activity toward single-stranded DNA than double-stranded DNA, and its catalytic activity against dU is stronger than that for dI and dX.

Mismatch cleavage; cleavage of oligonucleotides carrying modified bases including deoxyxanthosine (dX), deoxyinosine (dI), deoxyuridine (dU) and abasic (AP) sites, with higher cleavage activity toward dU than dI and dX; research on DNA repair.

Unit Definition
One unit is defined as the amount of enzyme required to cleave 1 pmol of a 60-mer single-stranded DNA oligonucleotide containing a single deoxyxanthosine site within a total reaction volume of 20 μL after incubation at 65 °C for 1 hour.
The deoxyxanthosine-containing substrate is a synthetic 5′ FAM-labeled 60-mer ssDNA oligonucleotide with dX incorporated at the 24th nucleotide position.
Purity
Free of all other DNA endonucleases, exonucleases and RNases apart from Thermostable Endonuclease Q.
Storage Conditions
Store at -20 °C, stable for at least 2 years.
