Streptavidin-FITC (Streptavidin-Fluorescein isothiocyanate), also known as FITC-Streptavidin, SA-FITC, FITC-SA, Fluorescein isothiocyanate-labeled Streptavidin, FITC-labeled Streptavidin, Streptavidin-Fluorescein isothiocyanate conjugate, FITC-conjugated Streptavidin, FITC-labeled Streptavidin, Streptavidin-fluorescein, Streptavidin, FITC Conjugate, Streptavidin, FITC conjugated, etc., can be used for the fluorescence detection of biotin-labeled antibodies, nucleic acids, proteins, or other biotin-labeled molecules. Specific applications include immunofluorescence, immunohistochemistry or immunocytochemistry, in situ hybridization, flow cytometry, etc.
The molecular weight of Streptavidin is 52 kD, and it can bind specifically to biotin with a high affinity. The affinity constant (Kd) between Streptavidin and biotin is approximately 10^-15 M. Streptavidin is a tetrameric protein capable of simultaneously binding four biotin molecules, originally purified from Streptomyces avidinii, and it shares high structural similarity and affinity for biotin with Avidin derived from chicken egg whites. Unlike Avidin, Streptavidin is a non-glycosylated protein and is essentially uncharged. Avidin has a pI (isoelectric point) of approximately 10.5, making it alkaline under neutral pH conditions. Due to the lack of charge under neutral conditions, Streptavidin exhibits much lower non-specific binding compared to Avidin, resulting in significantly reduced non-specific background in detection. Therefore, Streptavidin is commonly used instead of Avidin in biotin detection.
FITC is a commonly used green fluorescent probe that emits a green fluorescence signal upon excitation. It can be detected using fluorescence microscopy, fluorescence plate readers, or flow cytometers. The absorption (excitation) and emission peaks of FITC are as follows:
- Fluorophore: FITC
- Absorption Peak: 492nm
- Emission Peak: 520nm
This Streptavidin-FITC is prepared by coupling high-purity, high-quality Streptavidin with FITC, ensuring low background signals and high sensitivity.
The concentration of this Streptavidin-FITC is 1 mg/ml, and the recommended initial dilution is 1:50-200. During actual experimental procedures, the dilution ratio of Streptavidin-FITC may need to be adjusted according to specific conditions.
