Soybean Trypsin and Chymotrypsin Inhibitors (Premium Grade) contain two types of protein components derived from soybeans: Bowman-Birk-type soybean trypsin inhibitor (BBTI or BBI) and Kunitz-type soybean trypsin inhibitor (KTI). This product exhibits both trypsin inhibitory activity and chymotrypsin (also known as chymotrypsinogen) inhibitory activity. The Trypsin Inhibitor (TI) activity of this product is >15,000 BAEE units/mg, and the Chymotrypsin Inhibitor (CI) activity is >40 BTEE units/mg. It possesses extremely high purity and is suitable for applications such as biochemical assays, cell culture, and pharmaceutical excipients.
The product is prepared by mixing purified proteins of the two inhibitors with disodium hydrogen phosphate, monosodium phosphate, and purified water, followed by formulation, filtration, sterilization, and spray drying to obtain a powder. The powder ranges from white to brown and is readily soluble in water, with a neutral pH in aqueous solution.
Soybean Protease Inhibitor mainly acts as a serine protease inhibitor, capable of inhibiting proteases such as trypsin and chymotrypsin, among others. It is commonly used in serum-free cell culture, stem cell culture, pharmaceutical excipients, and other biochemical experiments requiring protease inhibition. This Soybean Trypsin and Chymotrypsin Inhibitors are extracted from non-genetically modified soybeans, ensuring high purity and no contamination with other proteases. The entire production process is free from animal-derived safety risks.
Trypsin Inhibitor (TI) and Chymotrypsin Inhibitor (CI) are essential biochemical reagents widely sourced from various origins. Soybeans are particularly rich in highly active trypsin and chymotrypsin inhibitors. Soybean contains two major types of protease inhibitors based on their amino acid composition, molecular weight, isoelectric point, affinity to different proteases, and immunological reactivity. The Bowman-Birk type soybean trypsin inhibitor (BBTI or BBI) is approximately 10kDa with 110 amino acids and exhibits inhibitory activity against both trypsin and chymotrypsin. The Kunitz type soybean trypsin inhibitor (KTI) is approximately 20kDa with 216 amino acids and primarily inhibits trypsin activity. Soybean Trypsin and Chymotrypsin Inhibitors effectively bind to proteases in a 1:1 molar ratio, forming stable complexes that block the active sites of the enzymes. Once this complex is formed, trypsin can cleave the arginine-isoleucine bond within the inhibitor. The inhibitory effect of soybean trypsin and chymotrypsin inhibitors on proteases is reversible and pH-dependent. The optimal pH range for inhibiting trypsin is pH 6.0-10.0, and it does not bind to proteases under pH < 5.0 conditions. Under the optimal pH value of 8.0, the association constant (Ka) between the inhibitor and trypsin is ≥10×108.
Under the conditions of 25°C and pH 7.6, using N-benzoyl-L-arginine ethyl ester (BAEE) or N-benzoyl-tyrosine ethyl ester (BTEE) as substrates, the enzyme quantity of trypsin or chymotrypsin that catalyzes the hydrolysis of 1 μmol of substrate per minute or produces an increase in ΔA253 of 0.001 per minute (reaction volume = 3.2 ml, path length = 1 cm) is defined as one trypsin unit or one chymotrypsin unit. The activity that can inhibit one trypsin or chymotrypsin unit is referred to as one unit (U) of inhibitory activity. For example, an activity of >9000 BAEE units/mg means that each milligram of the product can inhibit at least 9 mmol of trypsin that can hydrolyze BAEE under appropriate conditions.
- Before drying, this product undergoes sterile filtration. However, when used for cell culture, it is still recommended to prepare a solution and perform sterile filtration.
- This product is hygroscopic and should be stored in a dry place.
