Description
SMCC Activated Allophycocyanin (SMCC-APC), also known as SMCC Activated APC, is formed by modifying Allophycocyanin (APC) with Succinimidyl-4-(N-maleimidomethyl) cyclohexane-1-carboxylate (SMCC). This allows APC to conjugate with molecules containing thiol groups, such as proteins. The conjugates are commonly used in fluorescent microscopy detection, fluorescent immunoassays, dual or multi-color fluorescence analysis, cancer cell surface antigen detection, flow cytometry, antibody fluorescence labeling, in vivo imaging, and various diagnostic and bioengineering techniques.
APC is a phycobiliprotein extracted from spirulina (a type of blue-green algae) with a molecular weight of approximately 104 kDa. It has a very high quantum yield and absorption rate, producing intense and stable fluorescence. It easily crosslinks with other molecules, has low nonspecific adsorption, and is over 100 times more sensitive than conventional organic fluorophores, making it widely used as a fluorescent label in biological detection. The maximum excitation wavelength (Ex) of APC is 652 nm, and the maximum emission wavelength (Em) is 662 nm.
SMCC is a common amine-thiol crosslinker containing an amine-reactive N-hydroxysuccinimide (NHS) ester group and a thiol-reactive maleimide group. Under pH 7-9 conditions, the NHS ester group reacts with the primary amines of APC to form amide bonds, while the maleimide group can react with thiols on other molecules, such as proteins, under pH 6.5-7.5 conditions to form stable thioether bonds. The presence of cyclohexane in the SMCC structure significantly reduces the hydrolysis rate of the maleimide group, allowing SMCC-modified proteins to be stored in a lyophilized form.
This product is a conjugate of APC and SMCC, purified and free of excess SMCC, dissolved in PBS, and finally provided in a lyophilized powder form. It retains thiol reactivity even with long-term storage, ensuring optimal conjugation of SMCC-APC with other thiol-containing molecules such as proteins.
This product can be conjugated to biological macromolecules such as antibodies, which contain cysteine residues with thiol groups. When the thiol groups on cysteine form disulfide bonds, they can be treated with reducing agents to generate free thiol groups. When biological macromolecules such as proteins have available primary amine groups, the required thiol groups can also be added through chemical reactions.
Storage
Store at 4ºC protected from light, valid for one year. Do not store at -20ºC or lower temperatures. After preparing the stock solution with PBS, store at 4ºC protected from light and use within two weeks.
Precautions
- Fluorescent dyes are subject to quenching, so store and use them protected from light to slow down the quenching process.
- After preparing the solution, store it at 4ºC protected from light and do not freeze. The conjugate products should also be stored at 4ºC protected from light and not frozen.
- Avoid using buffers containing primary amines and thiols during the reaction to minimize unnecessary competitive reactions.
- This product is intended for scientific research use by trained professionals only. It is not for clinical diagnosis or treatment, food, or drug use, and should not be stored in residential areas.
