Description
Kex2 protease is a calcium-dependent serine protease derived from yeast. It specifically recognizes and cleaves peptide bonds at the carboxyl terminus of double basic amino acids such as Arg-Arg, Lys-Arg, and Pro-Arg. Unlike pancreatic proteases, Kex2 does not recognize and cleave peptide bonds at the carboxyl terminus of single basic amino acids such as arginine or lysine. In yeast, Kex2 protease is responsible for processing the precursors of killer toxin and α-factor. The activity of Kex2 protease is not inhibited by conventional serine protease inhibitors such as antipain, PMSF, or TPCK. The recombinant Kex2 protease is expressed and produced by Pichia pastoris and exhibits the same enzyme specificity as native Saccharomyces cerevisiae Kex2 enzyme. It exhibits optimal activity at pH 9.0 and is stable at pH 5.0-6.0.

- Recommended usage: Recommended reaction buffer: pH 7.0-9.0, 50 mM Tris-HCl, 2 mM Ca2+. If not used immediately after dissolution, it is recommended to dissolve the lyophilized powder in 20 mM (pH 5.2) NaAc-HAc, 2 mM Ca2+ buffer. The final concentration of the enzyme after dissolution is approximately 1-10 mg/mL. Divide into aliquots as needed and store at temperatures below -20°C. Note: The optimal reaction pH for Kex2 is pH 9.0, and the optimal pH for storage is 5.0-6.0.
- Stability: Stable as a lyophilized powder at 2-8°C, protected from light and moisture. After dissolution in a storage buffer (20 mM NaAc-HAc, pH 5.2, 2 mM Ca2+), it can be stored at temperatures below -20°C for 6 months. Repeated freezing and thawing reduces activity by approximately 10% after three cycles and 15% after five cycles.

Storage
