Description
Recombinant Human NQO1 Protein (EC 1.6.99.2), namely Recombinant Human NAD(P)H Quinone Oxidoreductase 1 (NQO1), is also known as NAD(P)H:(quinone acceptor) oxidoreductase, DT-diaphorase, NADH-menadione reductase and other aliases. It is a homodimeric FAD-dependent flavoprotein. This product uses NADH or NADPH as electron donors to catalyze the two-electron reduction of quinones, nitroaromatic compounds, azo dyes and other substrates.
One of the most well-studied functions of NQO1 is to protect organisms against the harmful oxidation and arylation induced by quinones. It directly reduces quinones to hydroquinones, eliminating electrophilic quinones while bypassing the formation of semiquinone radicals and reactive oxygen species, thereby exerting quinone detoxification effects. In addition, NQO1 acts as an endogenous CoQ reductase and plays a vital role in preventing lipid peroxidation.
The catalytic reduction of menadione by NQO1 and the corresponding activity assay principle are described as follows. First, NQO1 utilizes NAD(P)H as the electron donor to reduce oxidized menadione to reduced menadione, with concomitant production of NAD(P)⁺. Via electron coupling, WST-8 is reduced to orange-colored formazan, which exhibits a maximum absorbance peak near 450 nm. The amount of formazan generated is proportional to NQO1 activity; therefore, NQO1 activity can be detected with high sensitivity by measuring absorbance at 450 nm.
Applications
Exhibits potent antioxidant activity to combat oxidative stress and reduce susceptibility to human diseases. It acts as a selective "gatekeeper" in modulating proteasomal degradation of specific proteins. For example, NQO1 binds to and stabilizes the tumor suppressor p53, preventing its proteasomal degradation.

Activity Definition
One unit is defined as the amount of enzyme required to reduce 1 pmol of menadione with simultaneous reduction of WST-8 per minute at 37 °C in 50 mM Tris-HCl, pH 7.4.

Precautions
