Description
Recombinant Human B4GALT1 Protein (Y285L), also referred to as Recombinant Human β-1,4-Galactosyltransferase 1 (Y285L) or simply β-1,4-GalTase 1 Y285L, is a human extracellular catalytic domain of B4GALT1 (Globular catalytic domain, Leu126–Ser398) recombinantly expressed in E. coli. A point mutation substitutes tyrosine (Tyr, Y) at position 285 with leucine (Leu, L). The Y285L mutation remodels the conformation of the substrate-binding pocket, enabling the enzyme to specifically transfer N-azidoacetylgalactosamine (GalNAz) from UDP-N-azidoacetylgalactosamine (UDP-GalNAz) onto N-acetylglucosamine (GlcNAc) to generate non-reducing β1-4 linked GalNAz.
The azide group is capable of reacting with alkyne, DBCO (Dibenzocyclooctyne) or DIBO (Dibenzylcyclooctyne)-modified fluorophores (Cy3, Cy5, AF488, AF555, etc.), biotin, amine groups and other probes. Accordingly, this product can be applied to in vitro glycoprotein labelling, research on cell and tumour surface glycan remodelling, glycan microarray fabrication, targeted drug delivery and related studies. The protein bears an N-terminal His-tag, which can be removed by adsorption using anti-His antibody agarose resins, magnetic beads or nickel affinity columns.
Recombinant Human B4GALT1 Protein (Y285L) is the human orthologue corresponding to Recombinant Bovine β-1,4-Galactosyltransferase 1 (Y289L) (CAS 24-1-1060), and the two enzymes can be used interchangeably.
The β-1,4-Galactosyltransferase (β4GalT) family consists of glycosyltransferases belonging to type II membrane proteins. They exert critical functions in the biosynthesis of glycoproteins and glycolipids, and are widely involved in cell-cell interactions, cell adhesion, signal transduction and developmental processes. The β4GalT family contains seven members, grouped into four clusters based on sequence homology: β4GalT1 & β4GalT2; β4GalT3 & β4GalT4; β4GalT5 & β4GalT6; β4GalT7. Individual members differ in tissue distribution, substrate specificity and biological functions.

Applications | This product transfers GalNAz moieties onto GlcNAc residues of proteins and monoclonal antibodies. The azide-modified substrates can be further labelled via CuAAC (Copper-Catalysed Azide–Alkyne Cycloaddition) or SPAAC (Strain-Promoted Azide–Alkyne Cycloaddition). Compatible probes include Alkyne-Biotin, DBCO-Biotin, DIBO-Biotin, Alkyne-Cy3, DBCO-Cy3, DIBO-Cy3, Alkyne-amine, DBCO-amine and DIBO-amine. |

Precautions
- Recombinant Human B4GALT1 Protein (Y285L) displays markedly lower affinity for UDP-Galactose than for UDP-N-azidoacetylgalactosamine (UDP-GalNAz). Therefore, Recombinant Human B4GALT1 Protein (≥4 U/μg) is recommended when UDP-Galactose is utilized as the substrate.
- Each freeze-thaw cycle may cause partial inactivation of the protein. After preparing the stock solution to the target concentration for the first time, aliquot the protein and store at −20 °C or lower. Repeated freeze-thaw cycles should be avoided as far as possible.
