Description
RapidShield™ Western Blocker is the latest generation of fast and efficient Western blocking solution, which overall outperforms traditional blocking solutions based on BSA (bovine serum albumin), skim milk, casein, and similar products. It is primarily used for the blocking of PVDF (polyvinylidene difluoride) or nitrocellulose membranes in Western blot (WB) experiments. It can also be used for the dilution of primary and secondary antibodies in Western experiments.
RapidShield™ Western Blocker is fast and efficient. The blocking time typically requires only 5-15 minutes, and compared to traditional Western blocking solutions such as BSA, skim milk, casein, and similar fast-blocking solutions available internationally, it demonstrates a stronger signal-to-noise ratio.
RapidShield™ Western Blocker yields minimal background after blocking. This blocking solution does not contain serum or albumin, ensuring a high signal-to-noise ratio.
RapidShield™ Western Blocker exhibits excellent compatibility, allowing compatibility with horseradish peroxidase (HRP), alkaline phosphatase (AP), and biotin-labeled secondary antibodies. The product includes preservatives that do not affect the activity of HRP and AP, ensuring undisturbed detection with HRP or AP-conjugated secondary antibodies. Moreover, this product does not contain biotin, avoiding interference with biotin-based detection methods.
RapidShield™ Western Blocker offers convenience in usage. It does not require the addition of any extra reagents and can be directly applied for membrane blocking in Western blotting.
Based on the requirement of 5-10ml of RapidShield™ Western Blocker per membrane for blocking, a 100ml package of this product can be used to block 10-20 membranes, while a 500ml package can be used to block 50-100 membranes.
Precautions
- Typically, the blocking time for PVDF and NC membranes using this product is 5-15 minutes. For antibodies with high background, you can try extending the blocking time to 30-60 minutes. Additionally, if needed, overnight blocking at 4°C is also possible.
- Since there is no single blocking buffer that is suitable for all experimental systems, for specific experiments, it may be necessary to consider using other more appropriate blocking buffers based on the specific requirements.
- When handling PVDF and NC membranes, use flat-tipped forceps and gently grasp the edges, avoiding any scratches, creases, or indentations on the membrane surface.
- Once PVDF membranes are wetted and activated, they should be kept moist throughout the Western blotting process. Depending on the specific step in the Western blotting procedure, they can be placed in appropriate solutions such as Western transfer buffer or washing buffer. Failure to keep the PVDF membrane moist may result in abnormal background that is difficult to block.
- This product is intended for scientific research use by professionals only and should not be used for clinical diagnosis or treatment, food or drug purposes, or stored in regular residential areas.
- For your safety and health, please wear appropriate laboratory attire and disposable gloves while handling.
