Description
Rapid Blocking Buffer (TBS-T) quick-dissolve pellets are pale yellow to off-white pellets. Each pouch can be used to prepare 100 ml of blocking solution, making it convenient to use. Its primary ingredient is carefully selected casein protein component, and when prepared as a 1× working solution, the protein concentration is 5%. This product is intended for use in the antibody blocking step of Western Blot and ELISA, and it comes in a TBS-T buffer (containing Tween20 as a blocking agent) version.
Rapid Blocking Buffer (TBS-T) utilizes casein protein that is compatible with the vast majority of proteins, providing excellent reactivity and compatibility. It allows for rapid blocking within 15 minutes, reducing experimental time for users. This product is pre-mixed with TBS-T for ease of use, and the presence of Tween20 in many Western Blot detection reactions enhances blocking efficiency.

Protocol
Preparation Steps
1. Measure approximately 50 ml of distilled water and add it to a beaker. Place a magnetic stirrer inside the beaker.
2. Position the beaker on a magnetic stirrer and slowly add the entire contents of one pouch of rapid blocking buffer (TBS-T). Stir the solution until it completely dissolves.
3. Add distilled water to the solution prepared in step 2, bringing the total volume to 100 ml, resulting in a 1× concentration.
Blocking Procedure
1. After completing the transfer, place the transfer membrane into a hybridization box. Add 10~20 ml of 1× Rapid Blocking Buffer (TBS-T), ensuring it covers the surface of the membrane overloaded with proteins. Place the hybridization box on a flatbed shaker at room temperature and incubate for approximately 10 minutes.
Note: The blocking efficiency of this product is significantly better in 10 minutes compared to the traditional BSA blocking for 1 hour. For antibodies with a higher background, consider extending the blocking time to 30~60 minutes.
2. The blocked membrane can then be used for subsequent experiments such as primary antibody incubation.
