Description

Protein Endotoxin Removal Kit, also known as the Protein Sample Endotoxin Removal Kit or Protein Endotoxin Cleanup Kit, is a reagent kit that can efficiently remove over 99% of endotoxins from protein solution samples within 15 minutes. When applied to a protein sample with an initial endotoxin concentration of 100 EU/ml, the endotoxin content can be reduced to below 0.1 EU/ml, reaching a level considered free from endotoxin contamination. Therefore, this kit finds wide applications in the de-endotoxin treatment of samples involved in experiments with cells or animals.

This kit is specifically optimized for the removal of endotoxins from protein samples and can also be used for the removal of endotoxins from nucleic acid or small molecule solution samples, although the effectiveness in removing endotoxins from nucleic acid samples is slightly lower than from proteins.

Endotoxins, also known as lipopolysaccharides, are a major component of the cell wall in Gram-negative bacteria. They possess strong immunogenic activity and cytotoxicity, causing significant interference in experiments involving animal or cell studies. During the expression and purification process of recombinant proteins, a large amount of endotoxins is released when bacteria are lysed. These endotoxins have a strong binding affinity to proteins and cannot be effectively removed through conventional protein purification processes. Therefore, endotoxins are common contaminants in the production process of recombinant proteins.

Traditional methods for endotoxin removal, such as ion exchange chromatography, ultrafiltration, adsorption, hydrophobic chromatography, etc., often suffer from drawbacks such as low endotoxin removal efficiency, high cost, long processing times, complex operations, significant protein loss, and impact on the biological activity of proteins. These limitations greatly hinder the production of endotoxin-free recombinant proteins.

The Protein Endotoxin Removal Kit efficiently removes endotoxins, achieving a removal rate greater than 99% within 15 minutes for protein samples with endotoxin concentrations ranging from 100 EU/ml to 10,000,000 EU/ml.

Features

  • This kit has high endotoxin removal efficiency and is easy and quick to operate. Users only need to add the provided Endotoxin Removal Solution from this kit to the protein sample to be processed in a certain ratio. By incubating at a specific temperature and performing centrifugation, one round of endotoxin removal processing can be completed in as little as 10-15 minutes. The Endotoxin Removal Solution is a ready-to-use reagent, requiring no preparation or dilution steps and can be used directly. In practical tests, the endotoxin removal rate is >99% within the range of 100 EU to 10,000,000 EU, effectively reducing the endotoxin content in the sample by 1000-2000 times. After 1-3 rounds of endotoxin removal processing, the endotoxin content in the sample can be reduced by 1000-20000 times.
  • This kit does not affect the biological activity of proteins, and the loss of samples is minimal. The provided Endotoxin Removal Solution in this kit can efficiently remove endotoxins from protein samples while preserving the biological activity of proteins. The loss of protein samples is very small, typically only about 1-5% per processing. After processing, the residual amount of Endotoxin Removal Solution in the protein sample is very low, with no significant impact on most biological experiments.
  • This kit can handle a wide range of sample volumes. It is suitable for endotoxin removal processing of protein samples ranging from tens of microliters to tens of milliliters.
  • The components of this kit are comprehensive. The kit provides the Endotoxin Removal Solution A and B for endotoxin removal, as well as a color indicator for phase separation (Separation Indicator) between the aqueous and organic phases. Users can choose to use them according to experimental requirements.

 

Storage

Store at -20°C, valid for two years. Store at 4°C, valid for three months.

 

Precautions

  • This kit is not suitable for processing protein samples containing high concentrations of glycerol (≥10%). Glycerol can affect the phase separation of the aqueous and organic phases during the endotoxin removal process.
  • The concentration of the protein sample to be processed should generally not exceed 30 mg/ml. The specific highest protein concentration that can be applied may vary slightly depending on the properties of the protein. When the concentration is too high, it is advisable to try diluting the protein sample appropriately before using this kit for endotoxin removal.
  • The vessels used in the experiment must be endotoxin-free. If using conventional glassware, it must be pre-treated to remove potential exogenous endotoxins, commonly done by dry baking at 250°C for at least 60 minutes or using other suitable methods that do not interfere with bacterial endotoxin testing. If using plastic vessels, choose instruments labeled as endotoxin-free and non-interfering with the experiment.
  • All reagents and solutions used in the experiment must be endotoxin-free to avoid contamination from exogenous endotoxins.
  • Prevent microbial contamination during the experimental operation.
  • This product is for scientific research use by professionals only and is not intended for clinical diagnosis or treatment. It should not be used for food or drugs and should not be stored in ordinary residential areas.
