Description
Protein A Agarose (Fast Flow, for antibody purification) is also known as Protein A Resin and is primarily used for antibody purification. It can also be used for immunoprecipitation (IP) or co-immunoprecipitation (Co-IP). This Protein A Agarose has alkaline resistance and can tolerate 0.1-0.5M NaOH.
For immunoprecipitation or co-immunoprecipitation, Protein A Agarose (Fast Flow, for IP) can also be selected.
Protein A is a cell wall surface protein found in Staphylococcus aureus with a molecular weight of 42kDa. Protein G is an immunoglobulin-binding protein expressed by C-type or G-type streptococcal bacteria. Protein A and Protein G have similar functions and can specifically bind to mammalian immunoglobulins (Ig). The binding site is usually the Fc region of the immunoglobulin, but some data show that Protein A can also bind to the Fab region of the human VH3 family, and Protein G sometimes binds to the Fab region as well. At the same time, their binding abilities to different immunoglobulin subclasses vary. Recombinant Protein A and Protein G, appropriately modified, can be combined with agarose gel to be used for antibody purification or immunoprecipitation.
Protein A Agarose is suitable for the purification of human IgG1, IgG2, IgG4, mouse IgG2a, IgG2b, and rabbit IgG, etc.
The recombinant Protein A in this product specifically binds to the Fc region of most mammalian IgGs with a molecular weight of 14kDa. This recombinant Protein A has been engineered to have an alkaline-resistant structure while retaining only the amino acid sequence associated with binding to the IgG Fc region, removing any non-specific binding sequences outside the binding site. This effectively reduces non-specific binding. Each Protein A molecule can bind to 2 IgG molecules.
This product is formulated in a suitable solution that can be directly used for antibody purification and immunoprecipitation. Each milliliter contains a total volume of 0.25 ml of agarose beads (precipitate). The volume indicated on the product refers to the total volume of the suspension.
Features
- With a high IgG binding capacity and excellent stability, each milliliter of Protein A agarose beads (precipitate) can bind to over 50mg of human IgG. It exhibits good dynamic binding capacity at different retention times and flow rates.
- It also possesses outstanding alkaline resistance and ultra-high reusability. After 200 in-place washes with 0.1M NaOH, there is no significant decrease in binding capacity. Even after 100 in-place washes with 0.5M NaOH, it still retains 80% of its binding capacity.
- It demonstrates good pressure resistance using a rigid cross-linked 4% agarose matrix, with a maximum pressure tolerance index of 0.3MPa.
- It has an extremely low ligand leaching value, with a leaching value of less than 10ng/mg IgG, reducing the potential contamination caused by ligand leaching in downstream purification processes.
Precautions
- Do not freeze this product for storage.
- Before using Protein A Agarose, it must be thoroughly resuspended by gently inverting several times to ensure a uniform mixture.
- This product contains trace amounts of preservatives that do not interfere with routine antibody purification and immunoprecipitation. However, if subsequent enzymatic activity assays are involved, it is recommended to wash the Protein A Agarose beads three times with an appropriate solution such as TBS before use to eliminate any potential interference from the preservatives.
- Starting from the collection of protein samples, all steps involving protein samples must be performed at 4°C or on ice.
- This product is intended for scientific research use by professionals only and should not be used for clinical diagnosis or treatment, food or drug purposes, or stored in ordinary households.
