Description

The PrimeIAmp™ LAMP Lyophilized Isothermal Amplification Kit (pH Colorimetric) is based on a highly active Bst DNA Polymerase system, enabling rapid and efficient amplification of target nucleic acids under isothermal conditions. The kit is suitable for molecular diagnostics, point-of-care testing (POCT), field testing, and assay development.

This kit incorporates an aptamer-mediated hot-start technology. At low temperatures, the aptamer specifically binds to Bst DNA Polymerase and suppresses its polymerase activity, preventing unwanted amplification during reaction setup. As the reaction reaches the operating temperature, the aptamer undergoes a conformational change and dissociates from the enzyme, restoring polymerase activity and initiating amplification. This hot-start mechanism effectively minimizes non-specific amplification during reaction preparation and temperature ramp-up, thereby improving assay specificity, sensitivity, and reproducibility.

The kit is supplied in a convenient lyophilized, all-in-one format, with the essential amplification components and visual detection dye pre-mixed in each reaction. To perform the assay, users only need to add the sample, primers, and reaction buffer. Amplification results can be interpreted directly by visual color change without the need for specialized instruments, making the kit particularly suitable for rapid on-site testing, field surveillance, and decentralized laboratory applications.

A positive control is included for assay setup, workflow verification, and quality control, facilitating rapid assay development and routine testing. The kit offers high amplification efficiency, excellent sensitivity and specificity, outstanding reagent stability, and a streamlined workflow. It is well suited for applications including pathogen detection, genetic analysis, life science research, and POCT assay development.

Storage

Stable for 1 month at room temperature, 1 year at 4°C, and 2 years at −20°C.

 

Notes

  • It is recommended to include a no-template negative control in each experiment.
  • Prepare reagents and template DNA in separate areas to avoid contamination.
  • For optimal reproducibility, add the template DNA last.
  • Recommended amounts: plasmid DNA (10 fg–1 ng), genomic DNA (10 ng–100 ng). For optimization, adjust Mg²⁺ concentration (4–10 mM), Bst DNA Polymerase (0.04–0.32 U/µL), or reaction temperature (55–72°C).
  • Temperature control devices can include metal baths, water baths, or PCR machines. If using a PCR machine, ensure the heated lid is turned off before starting the program. Some PCR machines default to heating the lid to 105°C upon startup. It is recommended to run a few cycles to lower the lid temperature below 70°C.
  • Prepare reagents and templates in separate work areas to minimize the risk of contamination.
  • It is recommended to include a no-template control (NTC) in every experiment.
  • Temperature-controlled devices such as a dry bath, water bath, or PCR thermocycler may be used. On some PCR thermocyclers, the heated lid defaults to 105°C upon startup. In such cases, run several cycles or adjust the settings to reduce the heated lid temperature to 75°C before use.
  • To prevent evaporation of the reaction mixture, add 20 μL of mineral oil on top of the reaction mixture when using a water bath or dry bath. Mineral oil is not required when using a PCR thermocycler; however, the heated lid temperature must be set to 75°C.
  • This kit uses a pH indicator-based detection system and must be protected from exposure to air. Prolonged exposure may result in CO₂ absorption, causing reagent acidification and reduced assay accuracy. Storage: Do not store at room temperature or under refrigerated conditions (2–8°C). Store at −20°C.
  • For optimal reproducibility, add the template last when preparing the reaction mixture.
