Description
Precast PAGE Gel for Tris-Gly System is a new generation precast gel for coating plastic plates. It is a convenient, high-quality, and safe polyacrylamide gel with standard dimensions, designed for use with the Tris-Glycine electrophoresis buffer system. It allows for electrophoresis to be completed in just 40-60 minutes, producing very smooth and sharp bands. This precast gel includes a concentrated gel region of approximately 1.5 cm in height, providing excellent separation results. After electrophoresis, protein bands appear smooth, clear, fine, and sharp, with minimal edge effects. Additionally, this precast gel plate is made of coating plastic material, reducing non-specific protein adsorption, leading to superior electrophoresis performance, even surpassing that of self-made PAGE gels. It is commonly used for PAGE and Western blotting.
Precast PAGE Gel for Tris-Gly System is available in various gradient and fixed concentration gels, with options for 10-well and 15-well plates. Gradient gel concentrations range from 4-15%, 4-20%, 8-16%, and 8-20%, while fixed concentration gels are available in 6%, 8%, 10%, 12%, and 15%. The 4-20% gradient gel is the most commonly used. Customized services are available for special concentration requirements. Please refer to the table below for the optimal separation range of each precast gel:
Precast Gel Concentration | Number of Wells | Maximum Loading Volume | Electrophoresis/Transfer Buffer System | Optimal Separation Range |
---|---|---|---|---|
6% | 10/15 | 50/30μl | Tris-Glycine | 50-150kDa |
8% | 10/15 | 50/30μl | Tris-Glycine | 30-90kDa |
10% | 10/15 | 50/30μl | Tris-Glycine | 20-80kDa |
12% | 10/15 | 50/30μl | Tris-Glycine | 12-60kDa |
15% | 10/15 | 50/30μl | Tris-Glycine | 10-40kDa |
4-15% | 10/15 | 50/30μl | Tris-Glycine | 20-180kDa |
4-20% | 10/15 | 50/30μl | Tris-Glycine | 10-180kDa |
8-16% | 10/15 | 50/30μl | Tris-Glycine | 20-90kDa |
8-20% | 10/15 | 50/30μl | Tris-Glycine | 10-90kDa |
This precast gel contains approximately 1.5 cm of 4% concentrated gel, ensuring the production of very sharp bands effectively.
The ratio of acrylamide to bisacrylamide in this precast gel is 29:1, and the gel thickness is 1.0 mm. The maximum loading volume for 10-well configurations is 50 μl, while for 15-well configurations, it is 30 μl. The dimensions of the gel plate are 100 × 90 × 4.8 mm (width × height × thickness), and the gel size is 84 × 74 × 1 mm (width × height × thickness).
Polyacrylamide gel electrophoresis (PAGE) technology is widely used for protein separation, purification, detection, identification, and molecular weight analysis. It is one of the fundamental techniques in life science research. Common Western blot assays are based on PAGE.
The PAGE precast gel (Tris-Gly) is prepared with near-neutral pH Tris-HCl buffer, does not contain SDS, and can be used for both denaturing and non-denaturing protein electrophoresis.
Features
- Regarding the choice between 10-well and 15-well precast gels: When you have a larger number of samples to test or when quantification is required, it is recommended to use the 15-well precast gel. It offers greater throughput and is more convenient for quantitative statistical analysis of multiple samples. If you aim to obtain very high-quality representative images, it is recommended to use the 10-well precast gel, as it provides smoother and sharper bands.
- This product is safe and convenient. The precast gel requires no preparation and is ready to use; you can load your samples directly without the need for time-consuming gel preparation. This is in contrast to traditional PAGE gels, which involve handling toxic and irritating reagents during gel casting.
- This product offers stable quality. The use of coating plastic gel plates in the precast gel eliminates the risk of glass gel plates breaking during transport and usage, making it safer and reducing protein adsorption, resulting in better electrophoresis performance. The product is manufactured using a consistent production process, ensuring reliable and consistent quality across different batches.
- The electrophoresis results with this product are excellent. The precast gel provides exceptional protein separation, yielding smooth, clear, fine, and sharp protein bands, with high transfer efficiency.
- This product is compatible with a wide range of electrophoresis tanks available in the market.
- This product offers short electrophoresis times. The recommended electrophoresis voltage and time for this precast gel are 180V for 40-60 minutes or 150V for 60-70 minutes, resulting in smooth and sharp electrophoresis bands. Specific electrophoresis times can be adjusted based on the migration of bromophenol blue or specific experimental requirements.
- The stability and shelf life of PAGE precast gel (Tris-Gly) are good. The precast gel can be stored for up to 12 months when kept at 4°C. It retains its electrophoresis performance even after being stored at 37°C for 6 days, with no significant changes compared to storage at 4°C.
Storage
Store at 4°C, effective for one year. Do not freeze below 0°C.
