Description
Plant Tissue Optical Clearing Reagent is a product designed to render fixed plant tissues optically transparent. It effectively reduces autofluorescence from common pigments such as chlorophyll, while maintaining the stability of fluorescent proteins (FPs), thus enabling high-resolution imaging of deep plant tissues. This reagent can clear plant organs such as roots and leaves, or even entire plants, and is widely used in plant biology research for three-dimensional imaging. It is less suitable for highly lignified plant samples.
Plant organs and tissues contain various autofluorescent compounds like chlorophyll, which produce nonspecific background fluorescence during imaging, affecting observation quality. Moreover, plant tissues have structures with different refractive indices, causing light scattering and spherical aberration, leading to opacity and making deep tissue imaging very challenging. This reagent effectively removes common pigments like chlorophyll and adjusts the refractive index of plant tissues to match that of the solvent, minimizing light scattering and improving optical imaging quality.

- High Transparency: This product effectively processes plant samples to achieve high transparency. It has strong capabilities for removing chlorophyll and other pigments, significantly reducing autofluorescence in tissues. By adjusting the refractive index of plant tissues and organs to match the solvent, it increases light transmission, enabling effective transparency of plant tissues and organs.
- Wide Applicability: This product is suitable for rendering whole plants, tissues, and organs from various species transparent. It is also compatible with transgenic plants labeled with fluorescent proteins (FPs). Additionally, it works well with multiple chemical dyes, allowing for combined use to clear plant samples and facilitate subsequent staining and deep imaging.
- High Safety and Fluorescent Protein Stability: This product is non-toxic, free from traditional toxic clearing agents like chloral hydrate. It preserves the integrity of plant tissues and the fluorescence of proteins, making it safer to use.
Storage
- Store at room temperature for up to one year.
- Do not store at low temperatures to prevent precipitation.
Precautions
- When stored or transported at low temperatures, solid precipitation may occur. Place the product in a 37ºC water bath or incubator until the solution becomes clear before use. This does not affect its performance.
- The clearing process for this product takes longer. For plant materials or stains without special requirements, it is recommended to perform the clearing process first, then proceed with staining.
- This product can also be used to clear plant tissues after fluorescent staining without affecting the fluorescence. However, avoid light exposure to minimize fluorescence quenching.
- After clearing, delicate plant tissues may become soft. Use flat-tip tweezers carefully to handle samples to avoid damage.
- The amount of this product used should be sufficient to completely immerse the plant material and can be adjusted based on the sample size.
- This product is intended for use by professional researchers only. It is not for clinical diagnosis or treatment, nor for use in food or medicine. Do not store it in common household environments.
