Description
Phloxine B Paneth Cell Staining Kit, also known as Phloxine B Staining Kit or Paneth Cell Staining Kit, is a kit primarily using the dye Phloxine B to stain the eosinophilic granules in Paneth cells of the small intestine red, for the detection of Paneth cells. This kit is suitable for staining frozen or paraffin-embedded sections.
Paneth cells are highly specialized epithelial cells in the small intestine, located at the base of the intestinal glands, with dense secretory granules. These granules contain antimicrobial factors such as lysozyme (Lyz) and antimicrobial peptides (AMPs), which play a crucial role in maintaining intestinal homeostasis and defending against commensal and pathogenic microorganisms. Paneth cells also help maintain and regulate intestinal stem cells by secreting essential growth factors and regulatory molecules. Therefore, the absence or dysfunction of Paneth cells is associated with intestinal microbial imbalance, impaired tissue regeneration, and various inflammatory diseases.
Phloxine B, also known as Acid Red 92, is a fluorescent indicator and dye mainly used for biological staining and as a color additive in food, drugs, and cosmetics. When used in Gram staining, Phloxine B can differentiate between Gram-negative and Gram-positive bacteria. Orange G is an acid-base indicator and dye with a pH color change range of 11.5 (yellow) to 14.0 (orange-red), commonly used as a counterstain in Phloxine B staining.
This kit provides the main reagents needed for Phloxine B staining, including Phloxine B staining solution, Orange G counterstain solution, and hematoxylin staining solution for nuclear staining. The eosinophilic granules at the top of Paneth cells are easily stained by acidic dyes, and Phloxine B stains Paneth cells red with bright color. The subsequent use of Orange G counterstain solution ensures that the color of the granules is not easily faded during ethanol differentiation.
This kit is convenient to use, easy to operate, and provides clear staining results.
The staining solutions provided in this kit can be reused until the staining effect is deemed unsatisfactory, at which point new staining solutions should be used. The small packaging of this kit can stain at least 500 samples.

Storage
Store at room temperature, protected from light, valid for one year.
Precautions
- Differentiation after hematoxylin staining is an optional step, but it provides clearer nuclear staining.
- The staining solutions can be reused multiple times; replace them when the staining effect is deemed unsatisfactory.
- Paneth cell granules can be destroyed by acid. Fixation with neutral aldehyde solutions, such as 4% paraformaldehyde, is more effective. Fixation solutions containing potassium dichromate are better, while cells are easily destroyed when using Bouin’s solution, AAF, Carnoy’s solution, and other fixatives.
