Pepstatin A is a potent inhibitor of aspartic proteases, with inhibitory activity against targets such as hemoglobin-pepsin, hemoglobin-proctase, casein-pepsin, hemoglobinacid protease, and casein-proctase. In vitro studies demonstrate that Pepstatin A effectively inhibits various aspartic proteases, including those involved in HIV replication, without causing significant toxicity to infected cells. It also hinders osteoclast proliferation by blocking ERK signaling and suppressing NFATc1 expression. In vivo, Pepstatin A shows low toxicity levels, with LD50 values varying across species, and it effectively suppresses stomach ulceration in ligated Shay rats when administered orally.

- In Vitro Studies: Pepstatin A effectively inhibits aspartic proteases such as proteinase, tissue proteinase D, and E. In HIV-infected H9 cells, pepstatin A inhibits the synthesis of HIV gag protein within cells without significant toxicity to the infected cells. Pepstatin A inhibits osteoclast proliferation by blocking ERK signaling and suppressing the expression of NFATc1.
- In Vivo Studies: Pepstatin exhibits very low toxicity with LD50 values of 1090mg/kg, 875mg/kg, 820mg/kg, and 450mg/kg for mice, rats, rabbits, and dogs, respectively, when administered intraperitoneally (i.p.). It also shows LD50 values above 2000mg/kg for all species when administered orally. Pepstatin (0.5-50mg/kg, p.o.) suppresses stomach ulceration of the pylorus in ligated Shay rats.
