Description

Notopterol, a coumarin extracted from N. incisum, exhibits apoptotic properties alongside antipyretic, analgesic, and anti-inflammatory effects. It finds application in the treatment of acute myeloid leukemia (AML).

In Vitro Study

Notopterol induces cell apoptosis in a dose-dependent manner when administered at concentrations of 5, 10, 20, 40, and 60 μM for 48 hours. Additionally, it inhibits HL-60 cell proliferation in a concentration-dependent manner when applied at concentrations of 10, 20, 40, 60, and 80 μM for 24 to 120 hours. Moreover, Notopterol induces G0/G1 phase arrest in HL-60 cells when administered at concentrations of 5, 10, 20, and 40 μM for 48 hours, affecting cell-cycle proteins. Furthermore, it upregulates the expression of Bax and downregulates the expression of Bcl-2 and Mcl-1 in HL-60 cells after 48 hours of treatment with concentrations of 5, 10, 20, and 40 μM. Additionally, Notopterol promotes the cleavage of caspase 9, caspase 3, and PARP.
