In Vitro Study
Neohesperidin induces apoptosis in human breast adenocarcinoma MDA-MB-231 cells. The IC50 values of neohesperidin at 24 and 48 hours are 47.4±2.6 μM and 32.5±1.8 μM, respectively. Treatment with neohesperidin significantly up-regulates the expressions of P53 and Bax, while down-regulating that of Bcl-2 in the treated cells. Additionally, neohesperidin exhibits antioxidant activity with an IC50 value of 22.31 μg/ml in the DPPH radical-scavenging assay.
In Vivo Study
Neohesperidin administered at a dose of 50 mg/kg significantly inhibits 55.0% of HCl/ethanol-induced gastric lesions. In pylorus ligated rats, neohesperidin (50 mg/kg) significantly reduces the volume of gastric secretion and gastric acid output, while increasing the pH levels. Treatment with neohesperidin significantly lowers fasting glucose, serum glucose, and glycosylated serum protein (GSP) levels in mice. Furthermore, it markedly improves oral glucose tolerance and insulin sensitivity, while decreasing insulin resistance in diabetic mice. Neohesperidin also reduces serum triglycerides, total cholesterol, leptin levels, and liver index in these mice.
