Description
Multiplex STR PCR Master Mix (4X) is a highly convenient, 4× concentrated PCR premix specifically designed for multiplex amplification of genomic STR loci. This product contains a high-specificity Hot-Start Taq DNA Polymerase and a specially developed multiplex amplification buffer system, enabling efficient, specific, and sensitive simultaneous amplification of no fewer than 15 STR loci. It is particularly suitable for rapid and reliable STR genotyping of human, animal, plant, and bacterial genomic DNA.
The Hot-Start Taq DNA Polymerase used in this product is a mixture of Taq enzyme and a reversible inhibitor, allowing for efficient and convenient thermal activation. The inhibitor reversibly binds to the active site of Taq polymerase, forming an inactive complex at temperatures below 45ºC. This suppresses enzyme activity and effectively prevents non-specific amplification caused by primer-template misannealing or primer dimer formation at low temperatures. During subsequent PCR steps, when the temperature exceeds 45ºC, the inhibitor dissociates from the enzyme, releasing Taq polymerase activity only after the reaction enters the PCR phase. This ensures no DNA synthesis occurs before the initial denaturation step, significantly improving specificity, sensitivity, and quantitative accuracy.
Extensive testing has shown that, with optimized primers, this product supports simultaneous genotyping of at least 15 STR loci. In contrast, conventional PCR premixes often suffer from inconsistent amplification efficiency due to primer-template preferences and complexity, resulting in uneven amplification or non-specific products. This master mix incorporates a Hot-Start Taq DNA Polymerase and a repeatedly optimized buffer system tailored for STR multiplexing, ensuring balanced, high-efficiency, high-specificity, and high-sensitivity amplification across multiple target loci.
The master mix includes Hot-Start Taq DNA Polymerase, PCR buffer (with Mg²⁺), dNTPs, and stabilizers. Users only need to supply primers, template DNA, and deionized water—making it extremely convenient to use.
This product is compatible with STR genotyping of genomic DNA from humans, animals, plants, and bacteria. Simply add template DNA and primers, follow the recommended PCR protocol, and the resulting products can be directly analyzed via capillary electrophoresis or other genotyping methods.
When used in a 20 µl PCR reaction system, the small and medium package formats of this product are sufficient for approximately 50 and 200 reactions, respectively.

Store at –20ºC. Valid for one year.
Precautions
For multiplex fluorescent detection, primer design and concentration ratios must be properly optimized. Primers should be labeled with appropriate fluorescent dyes. STR multiplexing should only be performed after confirming optimal conditions.
Before use, ensure the reagent tube is fully thawed. Gently invert to mix—avoid generating bubbles during mixing.
Use filter tips when preparing the PCR reaction system to minimize contamination.
The product has been tested to withstand up to 10 freeze-thaw cycles without significant performance loss. However, repeated freeze-thawing should still be avoided to maintain optimal reagent quality.
Due to the high sensitivity of this product, it is strongly recommended to separate the following areas into three distinct rooms to prevent contamination:

Post-PCR product handling and electrophoresis
The area used for opening PCR tubes and performing capillary electrophoresis should be physically separated from the sample preparation and PCR setup areas.
