Description
Lambda Protein Phosphatase (recombinant λ protein phosphatase) is a manganese-dependent recombinant expressed and purified phosphoprotein phosphatase that can catalyze the dephosphorylation of serine, threonine, tyrosine, and histidine residues.
This product is a recombinant λ phosphatase with a 6×His tag, expressed in Escherichia coli. The phosphatase is encoded by the open reading frame ORF221 of the λ phage, which consists of 221 amino acid residues.
Lambda Protein Phosphatase can be used for dephosphorylation of proteins, allowing for the study of the relationship between protein phosphorylation, activity, and structure, as well as the validation of the specificity of protein phosphorylation site antibodies.
Unit definition of enzyme activity
Under the conditions of 30°C and pH 7.5, the amount of enzyme required to hydrolyze 1 nmol of p-nitrophenyl phosphate in 1 minute is defined as one unit of activity.
Protein Information
- Enzyme molecular weight: Approximately 26 kDa.
- Purity: Purity greater than 95%.
- The storage solution of Lambda Protein Phosphatase consists of 50 mM Tris-HCl (pH 7.5), 100 mM NaCl, 0.1 mM MnCl2, 0.1 mM EGTA, 2 mM dithiothreitol (DTT), 0.01% Brij35, and 50% (v/v) glycerol.
- The 10× Lambda Protein Phosphatase Buffer is composed of 500 mM Hepes (pH 7.5), 100 mM NaCl, 20 mM DTT, and 0.1% Brij35.
- The 10× MnCl2 solution is composed of 10 mM MnCl2.
- Inactivation: Lambda Protein Phosphatase can be inactivated by heating at 65°C for 1 hour in the presence of 50 mM EDTA.
Precautions
- Repeated freeze-thaw cycles can lead to a decrease in the activity of Lambda Protein Phosphatase. If storage at -80°C is desired, it is recommended to aliquot appropriately for convenient future use. It does not freeze when stored at -20°C, but it will freeze when stored at -80°C.
- 10 mM vanadate ions can inhibit approximately 90% of Lambda Protein Phosphatase activity, while 50 mM EDTA can inhibit approximately 95% of Lambda Protein Phosphatase activity. 1% Triton X-100, 0.4% Nonidet P-40, 0.025% Tween 20, 0.5 M NaCl, 0.1 mM ATP, 10 μg/ml pepstatin A, 10 μg/ml leupeptin, 10 μg/ml aprotinin, 0.5 mM PMSF, and 1 mM benzamidine do not affect the enzymatic activity of Lambda Protein Phosphatase.
- It should be noted that different protein dephosphorylation reactions have different rates. The optimal reaction temperature for this enzyme is 30°C. When using this enzyme for dephosphorylation treatment of crude extracts of certain samples, it is important to minimize the processing time and add an appropriate amount of protease inhibitors to reduce degradation of the target protein.

Storage
Store at -20°C, valid for one year. If longer storage is desired, it can be aliquoted appropriately and stored at -80°C.
