Description
Human Apurinic/apyrimidinic Endonuclease 1 (APE1), also known as APE1 endonuclease, HAP1 or Ref-1, is a human recombinant endonuclease homologous to Escherichia coli Exonuclease III and possesses AP lyase activity.
This product hydrolyzes the phosphodiester bond at the 5' side of apurinic/apyrimidinic (AP) sites, inducing single-strand DNA breaks and generating 3'-hydroxyl and 5'-deoxyribose phosphate (5'-dRP) termini. Meanwhile, it exhibits weak activities of DNA 3'-diesterase, 3'→5' exonuclease and RNase H. Beyond its DNA repair function, it can modulate the DNA-binding activity of a variety of transcription factors in vitro through a redox mechanism. It enhances the DNA-binding capacity of transcription factors, including Fos-Jun heterodimers, Jun-Jun homodimers, AP-1 protein derived from HeLa cells, NF-κB, Myb, as well as members of the ATF (Activating Transcription Factor)/CREB (cAMP Response Element-Binding Protein) family.

One unit is defined as the amount of enzyme required to cleave 20 pmol of a 34-mer double-stranded oligonucleotide containing one single AP site within a total reaction volume of 10 µL after incubation at 37 °C for 1 hour.
Purity
Free of other DNA endonucleases, exonucleases and RNases except APE1.
Inactivation and Inhibition
APE1 can be completely inactivated by heating at 65 °C for 20 minutes.
Storage Conditions
Store at -20 °C; stable for a minimum of 2 years.
