Description
Hematoxylin Staining Solution integrates various classic hematoxylin staining methods, such as the Harris method, Mayer method, etc. It simplifies the operation steps, shortens the operation time, and the staining solution does not contain toxic reagents such as mercury and methanol. It can be used for staining tissue sections or cultured cells.
Hematoxylin staining, also known as hematein staining or haematoxylin staining, is one of the most commonly used methods for staining tissues and cells. Colorless hematoxylin is oxidized to form oxidized hematoxylin (hematein or haematein), which has a quinoid ring structure. It can form colored complexes with trivalent aluminum ions, iron ions, etc., forming positively charged complexes like hematein-Al3+ complexes. The process of forming colored complexes between oxidized hematoxylin (also called oxidized hematein) and aluminum ions is also known as Dye Lake Formation. In the double-helix structure of genomic DNA in the cell nucleus, the phosphate groups on the double strands are outward-facing and negatively charged, making them acidic. They easily combine with positively charged complexes of oxidized hematoxylin, leading to nuclear staining. There are various methods for preparing hematoxylin staining solutions, and different methods can result in nuclear staining with varying shades of blue or bluish-purple.
After staining with this hematoxylin staining solution, cell nuclei appear blue.
This staining solution can be used in conjunction with immunofluorescence staining or immunohistochemistry staining. On one hand, you can perform immunofluorescence staining or staining with other dyes after staining with this hematoxylin staining solution. On the other hand, you can perform hematoxylin restaining after immunohistochemistry staining.
This staining solution can be reused until the staining effect is considered unsatisfactory, at which point you can replace it with a new staining solution. One 100ml package of this staining solution can stain at least 200 samples, while one 500ml package can stain at least 1000 samples.
Storage
Store at room temperature in a light-protected manner. It remains effective for at least one year.
Precaution
- Differentiation after staining is an optional step, but nuclear and cytoplasmic staining becomes clearer after differentiation.
- The staining solution can be reused multiple times; replace with new staining solution when the results are unsatisfactory.
- For first-time use of this kit, it is recommended to perform a pre-experiment with 1-2 samples.
