Description
Hematoxylin (also known as Hematein) is an organic dye that appears as a brown crystalline powder. It is commonly used for staining cell nuclei. Colorless Hematoxylin is oxidized to form Hematein or Haematein, which has a quinoid ring structure, and can form positively charged complexes with trivalent aluminum ions, iron ions, and other ions, resulting in colored compounds (such as hematein-Al3+ complexes). The process of forming colored complexes between oxidized Hematoxylin (also known as oxidized Hematein) and aluminum ions is also referred to as Dye Lake Formation. In the double-helix structure of genomic DNA within the cell nucleus, the phosphate groups on the double strands are negatively charged and acidic, which readily binds to positively charged complexes of Hematein, thereby facilitating cell nucleus staining. There are various preparation methods for Hematoxylin staining solutions, and these different methods can result in varying shades of blue or bluish-purple staining of cell nuclei.

Application
Hematoxylin staining, also known as Hematoxylin and Eosin (H&E) staining, is one of the most common methods for staining tissues and cell nuclei. It is often used in combination with Eosin staining, which stains the cytoplasm, for staining tissue sections or cultured cells [1]. There are various preparation methods for Hematoxylin staining solutions, and these different methods can result in varying shades of blue or bluish-purple staining of cell nuclei. Hematoxylin staining has been applied to the staining of gastric cancer tissues, cardiac tissues, heart sections, ovarian nuclei, and for diagnosing stages of tumor invasion. Furthermore, Hematoxylin is also utilized as an acid-base indicator. Through testing, this product has been shown to be effective for preparing Hematoxylin staining solutions and subsequent staining processes.
