Description
Heatstable Nucleoside diphosphokinase is a Nucleoside Diphosphate Kinase (NDPK) homologous protein isolated from Thermus thermophilus. This enzyme not only catalyzes the transfer of high-energy phosphate groups between adenosine triphosphate (ATP) and nucleoside diphosphate (NDP), but also has the activities of NDP kinase and protein phosphotransferase. It also participates in transcriptional regulation and signal transduction. Within the optimum temperature range of 60-75°C, it is responsible for the generation of ribonucleotides other than ATP in vivo, and the generation of triribonucleotides from dinucleotides: (d)XDP+(d)YTP → (d)XTP+(d)YDP.
Unit Definition
One unit is defined as the amount of enzyme required for converting 1.0 μmole TDP to TTP per minute in the presence of ATP at 37°C. The specific activity of the enzyme is 1X10e5 U/mg, and the concentration of the product is about 100 ng/ μl.
