Description
DNA/RNA Adenylase is SBS Genetech’s newly upgraded adenylation enzyme, engineered for high‑efficiency adenylation of 5′‑phosphate groups on ssDNA and ssRNA. It is recognized as the most efficient adenylation enzyme currently known. Under standard conditions, it can convert over 95% of phosphorylated DNA into adenylated DNA (AppDNA).
This enzyme is also extremely thermostable, retaining strong activity between 60–95 °C. High‑temperature adenylation not only promotes strand denaturation of DNA/RNA, thereby increasing the proportion of adenylated substrates, but also enables specialized workflows such as adenylation of fragmented DNA in NGS library preparation.
Contents
- DNA/RNA Adenylase (100 pmol/μl)
- 10×Adenylase Buffer
- 10 mM ATP
Note
- Enzyme-to-substrate ratio: During adenylylation, strictly control the molar ratio of enzyme to substrate. The ratio should be greater than 2, but not exceed 5. The recommended ratio is 2:1.
- Reaction temperature: The optimal reaction temperature is 75–80 °C. The enzyme remains active within 65–95 °C, and still retains >40% activity at 95 °C. Adjust as necessary.
- Magnesium dependence: The enzyme requires Mg²⁺ for activity, effective within the range of 2–10 mM.
- ATP dependence: The enzyme requires ATP for activity, effective within the range of 0.2–0.5 mM. Adjust as necessary. At concentrations below 0.1 mM, adenylylation efficiency drops sharply or may not occur.
- Substrate scope: The enzyme can adenylylate both ssDNA and ssRNA substrates with no difference in efficiency.
- Reaction pH: Since adenylylation products are unstable under high pH conditions (>8.5), the recommended reaction pH is 7.0–8.0.
- Evaporation control: Due to the high reaction temperature, evaporation may alter the system in small-volume experiments. To prevent this, overlay with mineral oil.
- Low substrate concentration: For adenylylation at low substrate concentrations, it is recommended to reduce the reaction volume rather than lowering the concentration of reactants, in order to achieve maximum efficiency.
Storage
The minimum shelf life is 3 years at -20°C.
