Description
DNA Methylation Analysis Kit (MspI/HpaII), also known as the DNA Methylation Analysis Kit (MspI/HpaII), is a simple, rapid, and efficient kit designed to analyze the methylation status of genomic DNA and other DNA samples based on the restriction enzymes MspI and HpaII.
DNA methylation is an important form of epigenetic modification. It mainly refers to the covalent bond formed between the 5th carbon atom of cytosine in genomic DNA and a methyl group, resulting in the modification of cytosine into 5‑methylcytosine (5mC). In eukaryotic genomic DNA, 5‑methylcytosine is the most common chemically modified base, and CpG dinucleotides are the primary methylation sites in the genome.
DNA methylation plays a crucial role in regulating gene expression, maintaining chromatin structure, genomic imprinting, X‑chromosome inactivation, and embryonic development. It also has profound effects on aging and the onset and progression of diseases. Numerous studies have demonstrated that DNA methylation is closely associated with various diseases, including cancer, psychiatric disorders, metabolic diseases, and autoimmune diseases. As a molecular biomarker or therapeutic target, DNA methylation can support precise disease diagnosis and treatment.
The DNA Methylation Detection Kit (MspI/HpaII) uses the restriction enzymes MspI and HpaII, which differ in their sensitivity to DNA methylation, to analyze the methylation status of specific sites. Both MspI and HpaII recognize the 5'-CCGG-3' sequence. However, when the internal CpG within the 5'-CCGG-3' tetranucleotide is methylated, HpaII digestion is blocked, whereas MspI digestion remains unaffected. Therefore, the MspI/HpaII combination enables analysis of the methylation level of genomic DNA at 5'-CCGG-3' sites.

Storage
Store at –20ºC. Stable for at least two years under recommended conditions.
Precautions
- The quality of DNA samples is critical for effective digestion by restriction enzymes.
- When using this product, keep it in an ice box or on ice. After use, immediately store it at −20ºC.
- When the cytosine outside the 5'-CCGG-3' site is methylated, digestion by both MspI and HpaII will be blocked.
- When the target gene contains 5‑hydroxymethylcytosine (5‑hmC), the digestion efficiency of MspI decreases significantly. Therefore, if the DNA sample is isolated from tissues rich in 5‑hmC (such as brain or stem cells), it is recommended to extend the digestion time to 4 hours.
