DNA-Free T4 DNA polymerase is a DNA polymerase derived from T4 bacteriophage, with a molecular weight of 104 kDa. It can catalyze the synthesis of deoxyribonucleic acid in the 5´→3’ direction in the presence of templates and primers. This enzyme possesses 3´→5´ exonuclease activity, which is 100-1000 times higher than that of E. coli DNA polymerase I Klenow fragment. It does not have 5´→3’ exonuclease activity.

Description

DNA-Free T4 DNA polymerase is a DNA polymerase derived from T4 bacteriophage, with a molecular weight of 104 kDa. It can catalyze the synthesis of deoxyribonucleic acid in the 5´→3’ direction in the presence of templates and primers. This enzyme possesses 3´→5´ exonuclease activity, which is 100-1000 times higher than that of E. coli DNA polymerase I Klenow fragment. It does not have 5´→3’ exonuclease activity.

Our DNA-Free T4 DNA polymerase is a recombinant protein and prepared through multiple purification steps, devoid of genomic and plasmid DNA from the production strain. 

 

Unit Definition

One unit is defined as the amount of enzyme required to incorporate 10 nmol dNTP into acid-insoluble material within 30 minutes at 37℃.

 

Features

  • Completely free from bacterial DNA.
  • No strand displacement activity.
  • No 5’-3’ exonuclease activity; cannot perform nick translation.
  • Synthesis error rate ~ 1×10⁻⁶ bases.
  • Heat inactivation: 75℃ for 20 minutes.

 

Applications

  • Gap filling (no nick translation activity).
  • Removal of 3’ overhangs or filling of 5’ overhangs to form blunt ends.
  • Creation of 5’ overhangs in seamless cloning for gene and vector complementary pairing.
