In Vitro Study
Columbianadin (CBN) effectively suppresses the growth of colon cancer cells. At low concentrations (up to 25 μM), Columbianadin induces apoptosis, while at high concentrations (50 μM), it induces necroptosis. The induction of apoptosis by Columbianadin involves modulation of caspase-9, caspase-3, Bax, Bcl-2, Bim, and Bid, while the induction of necroptosis is associated with RIP-3 and caspase-8. Additionally, Columbianadin leads to the accumulation of ROS and disrupts the balance of intracellular antioxidant enzymes such as SOD-1, SOD-2, catalase, and GPx-1. Columbianadin exhibits highly effective growth inhibitory activity against human colorectal cancer cells. Further investigation using HCT116 cells reveals the detailed growth-inhibitory mechanism of action mediated by Columbianadin. Treatment with various concentrations of Columbianadin (0-100 μM) results in dose- and time-dependent growth inhibition, with IC50 values of 47.2 μM and 32.4 μM after 48 and 72 hours of incubation, respectively. Treatment with Columbianadin at concentrations of 12.5, 25, and 50 μM for 48 hours in HCT116 cells decreases cell numbers and increases the proportion of floating cells. Morphological changes, including rounding and cell death, are observed particularly in cells treated with 25 and 50 μM Columbianadin.
In Vivo Study
The analysis method has been effectively applied to a tissue distribution study of Columbianadin (CBN) and Columbianetin (CBT) following intravenous administration of Columbianadin to rats. The findings of this investigation reveal that Columbianadin is detectable in all selected tissues after intravenous administration. Columbianadin is rapidly distributed to rat tissues and can undergo metabolism to CBT in most of the detected tissues. Notably, the heart exhibits the highest uptake of Columbianadin among the detected tissues, suggesting that the heart may be one of the primary target tissues of Columbianadin.
