Description
ClearVision™ Animal Tissue Transparency Kit is a highly effective and convenient kit that allows for superior optical clearing of animal bodies, whole organs, tissue blocks, and organ-like structures. This kit enables a quick and straightforward transparentization process, resulting in exceptional transparency. By utilizing this kit, animal samples can be seamlessly integrated with techniques such as immunofluorescence, immunohistochemistry, conventional or fluorescence staining, and bioluminescence. This integration allows for three-dimensional imaging of tissue blocks, organ-like structures, organs, and even entire animals. Consequently, the ClearVision™ Animal Tissue Transparency Kit finds extensive applications in the fields of biological and medical research.
CUBIC (Clear, unobstructed brain/body imaging cocktails and computational analysis) is a simple and effective method for achieving tissue optical clearing in animals. The ClearVision™ Animal Tissue Transparency Kit provides comparable transparentization effects to the conventional CUBIC method.
The ClearVision™-I Solution included in this kit not only facilitates the defatting of tissues and organs but also removes hemoglobin, thereby decolorizing them. Additionally, the ClearVision™-II Solution adjusts the refractive index of the tissue to match that of the solvent, effectively minimizing light scattering within the tissue and facilitating optimal optical imaging.
Features
- The experimental operation of this kit is simple and convenient. By simply immersing animal tissues, organs, and other samples in the reagents provided with this product, it is easy to achieve transparency without the need for any special instruments or equipment.
- The tissues and organs treated with this kit exhibit high transparency. Traditional tissue clearing methods focus on achieving a matched refractive index for individual tissues, which often results in reduced transparency. The ClearVision™-I Solution used in this kit has strong decolorization and defatting capabilities. When combined with the ClearVision™-II Solution, it effectively adjusts the refractive index of tissues and organs to match that of the solvent, increasing tissue transparency. The transparency achieved with this product is typically superior to methods such as SCALEVIEW-S, SeeDB2, or SeeDB. The transparentization effects of this product are comparable to Fujifilm/Wako's CUBIC Trial Kit, TCI's Tissue Clearing Reagent CUBIC-L [for Animals], and Tissue-Clearing Reagent CUBIC-R+(M) [for Animals]. The performance of this product is similar to Thermo's CytoVista™ Tissue Clearing Kit and CytoVista™ Tissue Clearing/Staining Kit, as well as Abcam's Tissue Clearing Kit.
- The clearing time with this kit is short. The traditional methods like SCALEVIEW-S or SeeDB2 usually require a longer time for tissue and organ clearing. Typically, the transparentization of mouse tissues and organs can be achieved within 1-3 weeks using this kit. The reference table below provides an estimate of the time required for transparentization of different tissues and organs in 6-week-old C57BL/6 mice. Actual results may vary depending on specific experimental conditions, so the table is for reference purposes only.
- The kit has wide applicability. It can be extensively used for the optical clearing of animal tissues, organs, and even the entire organism.
- The kit is compatible with fluorescent proteins and subsequent fluorescence staining. During the usage of this kit, it ensures the stability of fluorescent proteins and prevents the denaturation and loss of fluorescence in proteins such as EGFP, EYFP, mCherry, and others.
Precautions
- Before using ClearVision™-I Solution and ClearVision™-II Solution, allow them to reach room temperature and completely dissolve. Ensure that the liquids are transparent and free of precipitation. If crystals form or the solution becomes opaque, briefly place it in a 55°C water bath until it becomes completely transparent and free of precipitation.
- ClearVision™-I Solution and ClearVision™-II Solution are not effective at high temperatures (>75°C). Avoid subjecting them to high temperatures.
- Tissues may become soft after treatment with ClearVision™-I Solution. Use a spoon carefully for sampling and avoid using tweezers to prevent tissue damage.
- ClearVision™-I Solution can easily remove oily pen marks. Please make sure to tightly close the lid and seal it with Parafilm to prevent leakage. It is recommended to label the sample on both the cap and the tube to avoid losing sample information.
- If a strong ammonia odor is detected during the use of ClearVision™-I Solution, it indicates that the solution may have expired. Replace it with fresh ClearVision™-I Solution promptly to avoid compromising the transparency effect.
- When using ClearVision™-II Solution, avoid vigorous shaking to prevent the formation of bubbles. If a large number of bubbles accidentally form in ClearVision™-II Solution, they can be removed by using an ultrasonic cleaning machine or applying vacuum (~0.1 MPa, ~30 min) to eliminate the bubbles. Alternatively, allow the solution to sit at room temperature (for at least 7-8 hours) to allow the bubbles to dissipate naturally and prevent the occurrence of bubbles around or inside the tissue or organ.
- ClearVision™-II Solution is viscous. When using it, slowly draw it up with a wide-bore pipette tip (the tip can be cut with scissors), and slowly add it to the sample. Avoid vigorous pipetting to minimize bubble formation.
- After completing photo and video capture, the tissue or organ samples can have the Mounting Solution removed and then be immersed in ClearVision™-II Solution at room temperature for 1 week. The longer the immersion time, the higher the sample transparency, but it may also cause tissue expansion. After photo and video capture, the samples can be washed with 1X Wash Buffer, dehydrated with 30% (w/v) sucrose solution, embedded in O.C.T. compound, and stored long-term at -80°C.
- For thicker sections (>2 mm) or whole organs, the incubation times for each step can be extended according to the actual transparency effect.
- To ensure the long-term effectiveness of ClearVision™-I Solution and ClearVision™-II Solution, store them at -20°C in a light-protected manner. However, during usage, for convenient observation of tissue transparency, the samples can be processed under non-strict light-avoiding conditions.
- This product is intended for scientific research by professionals only and should not be used for clinical diagnosis or treatment, food or drug purposes, or stored in residential areas.
- For your safety and health, wear lab attire and disposable gloves when handling.
Storage
Store at -20°C, valid for one year. ClearVision™-I Solution and ClearVision™-II Solution should be stored at -20°C in a light-protected manner. The 100X Wash Buffer can be stored at 4°C. The Mounting Solution can be stored at room temperature or at 4°C.
