Description

Cell Species Identification Kit by Multiplex PCR, also known as the Cell Interspecific Cross-Contamination Detection PCR Kit, is a reagent kit that utilizes Multiplex PCR combined with agarose gel electrophoresis to rapidly and effectively perform common species identification and cross‑contamination detection. This kit can also be used for species identification of animal tissue samples (including hair). It enables simultaneous identification of five common species: human, African green monkey, Chinese hamster, mouse, and rat.

Cell species identification is a critical step in biomedical research, cell therapy, disease diagnosis, and the safety evaluation of biological products. Incorrect species attribution may lead to misinterpretation of research results. Commonly used methods for cell species identification include cell morphology analysis, karyotyping, isoenzyme analysis, DNA barcoding, STR profiling, and PCR‑based methods. Among these, STR profiling is widely used for human and certain rodent cell authentication and cross‑contamination detection; however, its applicability is limited to human and a subset of rodent species. Traditional karyotyping and isoenzyme analysis are also common tools, but they require large amounts of cells, involve complex procedures, have long turnaround times, incur high costs, and often lack sufficient sensitivity.

Features

  • This kit employs highly specific Multiplex PCR technology. It can simultaneously detect species‑specific DNA sequences from Homo sapiens (Human), Cercopithecus aethiops (African green monkey), Cricetulus griseus (Chinese hamster), Mus musculus (Mouse), and Rattus norvegicus (Rat). Validation studies show that the cross‑contamination detection limit reaches 1%, significantly improving detection efficiency and accuracy, and providing a rapid, reliable, and high‑performance solution for species identification in scientific research. The kit also includes a Positive Control to verify proper kit performance and corresponding band indication.
  • The kit is easy to use. It provides a rapid and convenient genomic DNA extraction reagent for cells or animal tissue samples, requiring no mechanical disruption, organic extraction, column purification, or DNA precipitation. The entire genomic DNA extraction process takes only about 20 minutes. The Multiplex PCR reaction mix included in the kit, Multiplex PCR Mix (2X, Green), contains 2X Loading Buffer and incorporates blue and orange electrophoresis tracking dyes (resulting in an overall green color). After PCR amplification, samples can be directly subjected to agarose gel electrophoresis without the need to add additional loading buffer.
  • The kit contains all necessary components. It provides all reagents required for cell species identification experiments, including the genomic DNA extraction reagent for cells or animal tissues, the Multiplex PCR Mix (2X, Green), a pre‑optimized Primer Pair Mix (10X), and a Positive Control, enabling users to conveniently perform sample testing and comparative analysis.

Components

  • DNA Extraction Solution
  • DNA Extraction Enzyme Mix
  • DNA Extraction Stop Solution
  • Multiplex PCR Mix (2X, Green)
  • Primer Pair Mix (10X)
  • Positive Control
  • Ultrapure Water

Storage

Store at –20ºC; stable for one year. The DNA Extraction Solution and DNA Extraction Stop Solution may be stored at 4ºC and remain stable for up to 3 months.

Precautions

  • The primer design of this kit fully considers both sequence conservation and specificity. However, even within the same species, slight variations in amplicon length may occur due to individual genetic differences, although the size will remain generally consistent with that of the positive control.
  • It is recommended to separate the sample preparation area, the PCR reaction setup area, and the post‑amplification analysis area into three independent rooms in sequential order to avoid product contamination. Areas where PCR tubes are opened and agarose gel electrophoresis is performed should be kept away from the sample preparation and PCR setup areas.
  • Validation tests show that up to 10 freeze–thaw cycles have no significant impact on kit performance; however, repeated freeze–thawing should still be avoided, as it may reduce product quality. If the Multiplex PCR Mix (2X, Green) is used frequently, an aliquot may be stored at 4ºC, where it remains stable for at least 7 days.
  • It is recommended to use filter tips when preparing PCR reaction mixtures to minimize the risk of contamination and false‑positive results.
  • Because PCR is extremely sensitive and can amplify target DNA sequences by more than ten million‑fold, care should be taken to avoid contamination with trace amounts of template DNA when using Taq polymerase. It is advisable to include a no‑template control to confirm the absence of contaminating DNA.
  • This product is intended for research use only by trained professionals. It is not for clinical diagnosis or therapy, not for use in food or pharmaceuticals, and must not be stored in residential settings.
  • For your safety and health, please wear a lab coat and disposable gloves during operation.
