Description
Calcium Colorimetric Assay Kit, also known as Calcium Assay Kit or Calcium Ion Assay Kit, is a colorimetric assay kit designed for the rapid quantitative detection of calcium ions in samples such as cells, tissues, serum, plasma, urine, and culture media.
This kit is user-friendly: liquid samples can be directly tested, and cell or tissue samples require only simple processing. The detection process is fast, taking as little as 10 minutes.
The kit has a wide linear range and high sensitivity: it shows a good linear relationship within the range of 0.2-2 µg of calcium ions and can detect as low as 0.1 µg of calcium ions.
Calcium is typically the most abundant mineral in mammalian bodies, involved in regulating numerous cellular activities and is one of the most important intracellular regulatory factors. Calcium can exist as free ions or in complexes with other ions, such as calcium phosphate and calcium carbonate, which are components of bone tissue. Many physiological processes, including muscle contraction, cell adhesion, hormone/neurotransmitter release, glycogen metabolism, cell proliferation/differentiation, blood coagulation, neural or synaptic transmission, and skeletal structure support, are regulated by calcium signaling. Defects in the integrity of cell-specific calcium signaling systems can lead to various diseases.
Although 99% of the body’s calcium is found in bones and teeth, clinical attention is often focused on blood calcium levels. Blood calcium levels are primarily regulated by parathyroid hormone, which adjusts calcium storage or release from bones to prevent hypercalcemia or hypocalcemia. Serum calcium exists mainly in three forms: about 50% as free calcium ions, about 45% as protein-bound calcium, and about 5% as calcium complexes (mainly calcium citrate). Calcium levels are usually inversely related to phosphorus levels.

Principle
The o-cresolphthalein complexone (OCPC) colorimetric method is a convenient, highly sensitive, and stable technique for calcium detection. Calcium ions react with o-cresolphthalein complexone under alkaline conditions to form a purple complex. The calcium ion content can be quantitatively detected by measuring the absorbance at 575 nm.
Storage
Store at -20ºC, valid for one year. Protect the color developing solution from light.
Precautions
- All samples should have a pH between 6 and 8. pH values that are too high or too low can affect the accuracy of the results. During sample addition and mixing, handle gently to avoid creating bubbles, as they may affect the final detection results. If the reaction temperature and time cannot be strictly controlled, a standard curve should be set up for each test.
- The detection buffer and color developing solution should be equilibrated to room temperature before use. The working solution, prepared by mixing the detection buffer and color developing solution, should be used immediately and is recommended to be used within 30 minutes.
- For plasma sample preparation, heparin can be used as an anticoagulant, but citrate, EDTA, and other substances that bind calcium ions should not be used. Chelating agents (such as EDTA, EGTA), citrates, oxalates, sodium fluoride, and other fluorides that can bind calcium ions will interfere with the detection of this kit.
- For samples with calcium ion concentrations that are too high or too low and fall outside the standard curve range, adjust the sample volume appropriately to bring the calcium ion concentration within the linear range of the standard curve.
- The detection buffer and color developing solution are harmful to humans. Handle with care and use effective protection to avoid direct contact with the body or inhalation.
